陇南核桃腐烂病一株伴生细菌的分离与鉴定

摘　　要：从甘肃陇南成县发现的核桃腐烂病病斑处,用组织分离法分离到一株腐生细菌菌株HBG.通过培养性状、形态特征、生理生化特征和16S rDNA序列分析进行鉴定.结果表明,该细菌为革蓝氏阴性菌G-细菌,有运动性.生理生化研究表明,该细菌接触酶、V-P试验、甲基红及糖类试验等均表现出阳性反应.最适生长温度为25~30℃,最适生长pH值为6.0~8.0.16S rDNA基因序列分析结果表明,该菌株的16S rDNA基因序列同源性最高的菌株为泛菌(Pantoea sp.),将该菌株鉴定为Pantoea sp.HBG..A pathogen,HBG,was isolated from the canker disease of walnut from Cheng county in Longnan,Gansu Province.The pathgen was identified according to the culture traits,morphological characteristics,physiological and biochemical traits and 16S rDNA sequence analysis.The result shows that this pathgen is a motile bacteria and it shows Gram negative by Gram staining.The catalase,V-P,methylene red and sugar fermentation positive.Its temperature optima is between 25℃ and 30℃,and pH optimum ranges from pH 6 to 8.Based on the 16S rDNA sequence analysis,the pathgen was identified to be Pantoea sp.HBG.
