摘  要:四环素具有残留量较高、危害性较强等问题。为了解决养殖场粪便中残留的四环素,试验以养殖场新鲜粪便为菌源样品,通过梯度选择及液相定量检测进行降解四环素菌株的筛选,对得到的菌株进行种属鉴定,然后考察pH值、无机盐、处理时间、菌量和溶氧等因素对其降解能力的影响。结果显示:通过筛选得到降解菌株N2-13,其对四环素的降解率达73.88%;经过形态鉴定、生理生化试验和16S rDNA序列分析,该菌为解淀粉芽孢杆菌(Bacillus amyloliquefaciens);该菌株在pH值7.0、装液量100 mL/250 mL、添加0.03%FeSO4、接种量2%、37℃培养3 d条件下降解率最高,达到97.27%;堆肥试验表明,添加N2-13降解菌后,四环素降解率达到94.51%。说明该菌株具有较好的应用潜力。研究扩大了四环素微生物降解的菌种资源,并为该菌株的实际应用提供参考。Because of the problems of high residual amount and strong harmfulness of tetracycline,in order to solve the residual tetracycline in the manure of the farm,taking the fresh feces of the farm as the bacterial source samples,the tetracycline-degrading strains were screened by gradient selection and liquid phase quantitative detection,and the species of the degraded strains were identified.Then the effects of pH value,inorganic salt,treatment time,amount of bacteria and dissolved oxygen on its degradation ability were investigated.The results showed that the degradation strain N2-13 was obtained by screening,and its degradation rate to tetracycline was 73.88%.After morphological identification,physiological and biochemical tests and 16S rDNA sequence analysis,the strain was Bacillus amyloliquefaciens.The strain was cultured at pH value 7.0,liquid volume of 100 mL/250 mL,added with 0.03%FeSO4,inoculated at 2%,and at 37℃for 3 d.Under the conditions,the degradation rate reached 97.27%.Compost tests showed that after adding N2-13 degrading bacteria,the degradation rate of tetracycline reached 94.51%,indicating that the strain has good application potential.The study expanded the resources of strains degraded by tetracycline microorganisms,and provided data support for the practical application of the strain.
