摘 要:研究对黄芩苷微粉添加剂进行急性毒性试验、生理生化、免疫指标和亚慢性毒性试验,评价黄芩苷微粉添加剂的安全性。结果表明:黄芩苷微粉添加剂经口半数致死量(LD50)为4.41 g/kg,LD50的95%可信限为3.21~6.04 g/kg。给药14 d后家兔血液中白细胞计数(WBC)和红细胞总数(RBC)下降,而肌酐(CRE)、尿素氮(BUN)、谷草转氨酶(AST)、谷丙转氨酶(ALT)有升高趋势,但差异不显著(P<0.05)。和对照组相比,给药第7 d和第14 d,中剂量组家兔血清免疫球蛋白A(IgA)、免疫球蛋白G(IgG)含量显著提高(P<0.05)。组织学结果表明,低剂量和中剂量组肝脏和肾脏组织结构清晰,高剂量组为肝索排列紊乱,肝窦、肾小球有少量出血。结果提示,常规剂量添加黄芩苷微粉添加剂安全性好,且能够提高动物机体免疫力,具有替代抗生素的潜在价值,具有较好的应用前景。Using the baicalin micro-powder,the acute toxicity test and chronic toxicity test were to evaluate the clinical safety of baicalin micropowder.The results showed that the acute toxicity test indicated that the LD50 of baicalin micropowder was 4.41 g/kg after oral administration of its suspension and 95%confidence interval was 3.21~6.04 g/kg.Hematology analysis showed that WBC and RBC count of rabbits decreased and CRE,BUN,AST of rabbits and ALT had elevated trend during 14 d after oral administration,but had no significant different(P>0.05).The contents of IgA,IgG of rabbits in medium-dose groups were significantly increased on 7 d and 14 d(P<0.05).There was no visible pathological changes on the liver,kidney histology.According to pathological observation the structure of liver and kidney were clear in low and middle dose group,but disorganized hepatic cell cords was found in the liver and hemorrhage between glomerulo and hepatic sinusoids in high dose group.In summary,the baicalin micro-powder could be considered as a safe in clinical treatment,and it can significantly improve the immunity.Baicalin has the potential to replace feeding antibiotics and has a good application prospect and promotion value.
