摘 要:目的探究慢性丙型肝炎病毒(Hepatitis C virus,HCV)感染通过转化生长因子-β(Transforming growth factor-β,TGF-β)对肝癌细胞侵袭性的影响。方法选取2016年6月-2018年6月于山西医科大学附属山西白求恩医院就诊的肝癌患者68例,根据其是否被HCV感染,分为感染组(n=21)和对照组(n=47)。采用实时荧光定量法对血清TGF-β进行检测;通过体外细胞侵袭实验和迁移实验分别研究TGF-β对HepG2细胞细胞侵袭、迁移能力的影响。结果感染组患者血清TGF-β水平为(461.72±24.96)mg/L高于对照组(198.54±18.10)mg/L(P<0.001);感染组的侵袭细胞个数为(97.69±14.52)个多于对照组(43.28±8.95)个(P<0.001);感染组的迁移细胞个数为(109.85±13.64)个为多于对照组的(51.30±8.27)个(P<0.001)。结论HCV病毒感染通过TGF-β促进肝癌细胞的侵袭性。OBJECTIVE To explore the effects of chronic hepatitis C virus(HCV)infection on invasiveness of liver cancer cells through transforming growth factor-β(TGF-β).METHODS A total of 68 patients with hepatocellular carcinoma(HCC)who were treated in Shanxi Bethune Hospital Affiliated to Shanxi Medical University from Jun.2016 to Jun.2018 were selected and divided into infection group(n=21)and control group(n=47)according to presence or absence of HCV infection.The serum TGF-βwas detected by real-time fluorescence quantification methods.The effects of TGF-βon invasion and migration of HepG2 cells were studied by in vitro cell invasion and migration experiments.RESULTS The level of serum TGF-βin infection group was(461.72±24.96)mg/L,sigificently higher than that in control group(198.54±18.10 mg/L)(P<0.001).The number of invaded cells in the infection group was(97.69±14.52),sigificently more than that in control group(43.28±8.95)(P<0.001).And the number of migrating cells in the infection group was(109.85±13.64),sigificently more than that in control group(51.30±8.27)](P<0.001).CONCLUSION HCV infection can promote invasiveness of liver cancer cells through TGF-β.
