摘  要:本试验对患蹄叶炎的奶牛通过蹄部注射牛脂肪间充质干细胞(bovine adipose-derived mesenchymal stem cells,bAD-MSCs)制剂进行疗效观察。将原代bAD-MSCs复苏传代并收集细胞与上清液,进行细胞超声破碎浓缩制作成bAD-MSCs制剂;利用小鼠和家兔对bAD-MSCs制剂进行安全性试验,然后对奶牛进行临床治疗试验,通过监测奶牛的体温、呼吸数以及跛行评分变化等评价其临床疗效。bAd-MSCs制剂对小鼠体重与主要脏器无影响(P>0.05);家兔体温升高均低于0.6℃,并且3只家兔体温升高总和低于1.3℃。在奶牛在体试验中,治疗组与对照组相比,试验前后的呼吸数与体温变化无显著差异(P>0.05)。但在跛行程度上,给药6 h后治疗组奶牛的跛行程度开始变轻(表现出治疗效果),在注射后48 h奶牛的跛行评分在治疗组和对照组分别是3.25±0.25和4.5±0.29,治疗效果明显(P<0.05);而在注射后(14 d)奶牛的跛行评分在治疗组和对照组分别是1.75±0.25和4.75±0.25,其治疗效果表现出极显著差异(P<0.001)。结果表明:bAD-MSCs制剂是无毒性、无热原性和无刺激性的制剂,患病奶牛在注射该制剂后48 h显著地降低了跛行评分,对奶牛蹄叶炎具有较好的治疗效果。本试验为防治奶牛蹄叶炎提供了新的研究方向。This study was performed to observe the therapeutic effect of bovine adipose-derived mesenchymal stem cells(bAD-MSCs)preparation on dairy cows with laminitis.First,the primary bAD-MSCs were thawed and sub-cultured,and the cells and supernatant were collected,then the cells were ultrasonically crushed and concentrated to form bAD-MSCs preparation.Secondly,safety analysis tests of the bAD-MSCs preparation were carried out in mice and rabbits.Finally,experiments of clinical treatment on dairy cows with laminitis were performed.The clinical efficacy was evaluated by monitoring the temperature,respiratory number and claudication score of the cows.The results were as follow:The bAd-MSCs preparation had no effect on the body weight and main organs of the mice(P>0.05).The body temperature rise of the rabbits was lower than 0.6℃,and their total body temperature rise was lower than 1.3℃.In the in vivo experiments with the cows,there was no significant difference in the respiratory number and body temperature between the treatment group and the control group before and after the experiment(P>0.05).However,the claudication degree of the cows in the treatment group became lighter after 6 h of administration,and the claudication scores of cows in the treatment group and the control group were 3.25±0.25 and 4.5±0.29,respectively,at 48 h after injection,with significant therapeutic effect(P<0.05).At 336 h after injection,the claudication scores was 1.75±0.25 in the treatment group and 4.75±0.25 in the control group,respectively,indicating an extremely significant difference in the therapeutic effect(P<0.001).These results suggested that bA D-MSCs preparation was non-toxic,non-pyrogenic and non-irritating,having a desirable therapeutic effect on laminitis in cows;which provided a new direction of research on the prevention and treatment of dairy cows with laminitis.
