摘 要:为提高鸡心包积液-肝炎综合征(hydropericardium hepatitis syndrome,HHS)和鸡包涵体肝炎(inclusion body hepatistis,IBH)疾病的防控能力,研制有效防控Ⅰ群禽腺病毒(FAdV)疫苗。本研究将实验室分离鉴定的FAdV-4 SN2016株和FAdV-8标准毒株,采用鸡胚增殖方式,优化最佳接种剂量、收获时间及甲醛的灭活条件,病毒抗原以3∶1比例混合,制备双价油乳剂灭活疫苗,检验疫苗性状、监测免疫后雏鸡的抗体水平及对其免疫保护效果。病毒采用卵黄囊接种,尿囊液中病毒粒子含量分别达到4.6×10^8和7.8×10^7拷贝/mL;灭活条件为终浓度1.5‰的甲醛灭活24 h;制备的疫苗为油包水剂型,物理性状稳定,无菌检测合格,4℃保存期可达12个月;雏鸡免疫抗体水平监测结果显示,免疫后3~4周抗体水平达到高峰,第6周依然维持较高水平,制备的双价油乳剂灭活疫苗可以有效抵抗单病毒和双病毒的攻击。研究结果显示制备的FAdV-4 SN2016和FAdV-8双价油乳剂灭活疫苗性状稳定、保存期长,免疫效果好,能有效抵抗病毒攻击,为HHS和IBH疫病的防控提供有效手段。The experiment was conducted to development vaccine of fowl adenovirus group-Ⅰ,in order to improve the prevention ability of hydropericardium hepatitis syndrome(HHS)and of inclusion body hepatistis(IBH).In this research,the FAdV-4 SN2016 strain previously isolated by our laboratory and FAdV-8 standard strain are proliferated by chicken embryo,the best inoculation dose,harvest time and formaldehyde inactivation conditions were optimized.Mixing virus antigens as the ratio of 3∶1,producted the bivalent oil-emulsion inactivated vaccine.The vaccine characters were investigated,immune antibody level was monitored and immunoprotective effect test was tested.Inoculated in yolk sac,the virion content reached 4.6×10^8 and 7.8×10^7 copies/mL respectively.The viruses were inactivated by formaldehyde with a final concentration of 1.5‰lasting 24 h.The vaccine showed white oil-emulsion and could be preserved 12 months at 4℃.The antibody level reached the OD450 value peak between week 3 and week 4 after immunization and remained higher even after 6 weeks.The vaccine showed 10^0%protection rate,and could effectively resist the attacks of monovirus and divirus.All the results proved that the FAdV-4 SN2016 and FAdV-8 bivalent oil-emulsion inactivated vaccine has long protection duration,good immune effects,and can very effectively resist infection of the corresponding virus.
