摘 要:以鹅为研究对象,人工感染禽腺病毒(fowl adenovirus,FAdV),分别在攻毒后36,72 h和3,7 d,采集脾脏、哈德氏腺、骨髓、法氏囊和盲肠扁桃体,运用实时荧光定量PCR,检测鹅感染FAdV-4后,免疫器官中FAdV-4载量及免疫相关因子基因表达差异,初探FAdV-4对鹅致病性以及鹅对该病毒的天然免疫反应机制。结果表明,与对照组相比,在法氏囊、盲肠扁桃体和哈德氏腺中检测到FAdV-4,感染初期TLR2和TLR3显著上调,感染后期TLR5和TLR15显著下调;AvBD1、AvBD9、AvBD10和AvBD16在部分组织中表达量显著升高,且大部分细胞因子表达受到抑制。研究表明,感染FAdV-4后,会激活宿主先天免疫反应,为研究FAdV-4与宿主先天免疫的相互作用提供新的思路。In this experiment the spleen,Harderian gland,bone marrow,bursa of Fabricius and cecal tonsil were collected at 36,72 h,3 and 7 d post infection(hpi or dpi).When the goose was infected with FAdV-4,differential expression of FAdV-4 load and immune-related factors in immune organs were detected by RT-qPCR.And the pathogenicity of FAdV-4 to goose and the natural immune response mechanism of goose to the virus were investigated.The results showed that FAdV-4 was detected in the bursa of Fabricius,cecal tonsil and Harderian glands,and TLR2 and TLR3 were significantly up-regulated at the early stage of infection,and TLR5 and TLR15 were significantly down-regulated at the late stage of infection;the expression levels of AvBD1,AvBD9,AvBD10 and AvBD16 were significantly increased in some tissues;and most of the cytokine expression were inhibited compared with the control group.The results showed that the host innate immune response was activated after infection with FAdV-4,The present funding provided new insights into the interaction between FAdV-4 and host innate immunity.
