摘 要:为了研究三氧化二砷(As2O3)对雄性小鼠生殖系统的影响,将40只6周龄雄性小鼠随机分为4组,以0,3,6,9 mg/kg的剂量连续4周皮下注射As2O3溶液,观察睾丸和附睾病理变化,检测精子活力、血浆和睾丸中砷含量、睾酮含量、抗氧化指标(MDA、GSH-Px)、生殖相关标志性分子AR、GPx5和Miwi基因表达以及Miwi定位表达。结果显示,As2O3处理组中睾丸生精管畸形,管内生精细胞排列紊乱,附睾管内精子和生精细胞显著减少。随着As2O3浓度的增加,精子活力下降,血浆和睾丸中的砷含量增加,睾酮水平降低。在血浆和睾丸中,MDA和GSH-Px含量在6和9 mg/kg As2O3组显著增加(P<0.05)。生殖相关标志性分子AR、GPx5和Miwi的表达量与对照组相比显著降低(P<0.05)。Miwi在睾丸初级精母细胞中的定位表达随着As2O3剂量的增加明显降低。上述结果表明,As2O3诱导小鼠发生氧化应激,阻碍了小鼠的精子成熟和精子发生,导致生殖毒性。To investigate the effects of arsenic trioxide(As2O3)on the reproductive system of male mice,forty 6-week-old male mice were randomly divided into 4 groups and subcutaneously injected with As2O3 solution at the doses of 0,3,6 and 9 mg/kg for 4 weeks.The pathological changes of testis and epididymis were observed.The sperm motility,the content of arsenic and testosterone in plasma and testis,antioxidant indexes(MDA,GSH-Px),the expression of AR,GPx5 and Miwi,the localized expression of Miwi were detected.The results showed that the testicular spermatogenic duct malformation,the arrangement of spermatogenic cells in the tube was disordered,and the sperm and spermatogenic cells in the epididymal tube decreased significantly with the increase of As2O3 concentration.The sperm motility and the level of testosterone decreased,the content of arsenic increased.The contents of MDA and GSH-Px were increased significantly in 6 and 9 mg/kg As2O3 groups(P<0.05).The expression of AR,GPx5 and Miwi were down-regulated significantly compared with control group(P<0.05).The localized expression of Miwi in testicular primary spermatocytes decreased.These results suggested that As2O3 induced oxidative stress in mice,hindered sperm maturation and spermatogenesis and lead to reproductive toxicity.
