摘  要:目的观察槐耳颗粒对肝癌侧群(side population,SP)细胞阿霉素耐药性的影响。方法采用流式细胞术分离肝癌细胞株MHCC-97L的SP细胞和非SP细胞,分选后的细胞分别经阿霉素(500 ng·mL-1)处理。采用不同质量浓度非细胞毒性剂量(0、2、4、8 mg·mL-1)的槐耳来进行分组实验。MTT法测定细胞增殖活性,并计算细胞抑制率,Western blotting检测耐药转运蛋白P-gp、ABCG2表达水平。结果SP细胞集落形成能力明显强于非SP细胞,形成集落数分别为5.3±1.9、2.2±1.5(P<0.01);槐耳能抑制SP细胞增殖,在质量浓度为10和12 mg·mL-1时更为明显(P<0.05),且在质量浓度为4和8 mg·mL-1时即可增强阿霉素对SP的增殖抑制作用(P<0.05);槐耳能减少耐药转运蛋白P-gp(P<0.05)和ABCG2(P<0.01)的表达。结论槐耳能减少并逆转肝癌SP细胞对阿霉素的耐药性。Objective To observe the effect of Huaier granule on doxorubicin resistance of hepatoma side population(SP)cells.Methods The SP and non-SP cells were isolated from MHCC-97L cell line by flow cytometry,and were treated with doxorubicin(500 ng·mL-1).Furthermore,cells were supplied with non-cytotoxic doses of Huaier(0,2,4,8 mg·mL-1).Cell proliferation was measured by MTT assay to calculate the cell inhibition rate.Western blotting was used to detect the expression of drug resistance transport protein P-gp and ABCG2.Results The number of colonies formed in SP cells(5.3±1.9)was higher than that in non-SP cells(2.2±1.5)(P<0.01).Supplement with Huaier inhibited the proliferation of SP cells at concentrations of 10 and 12 mg·mL-1(P<0.05),and enhanced the inhibition of doxorubicin on SP cells at 4 and 8 mg·mL-1(P<0.05).In addition,Huaier treatment reduced the expression of P-gp(P<0.05)and ABCG2(P<0.01).Conclusion Huaier can reduce and reverse the resistance of hepatoma SP cells to doxorubicin.
