摘 要:目的探讨乌梅等8种中药颗粒剂对泛耐药鲍曼不动杆菌的体外抑菌机制。方法采用试管稀释法测定乌梅、五味子、薄荷、赤芍、黄连、艾叶、连翘、黄芩颗粒剂悬液对泛耐药鲍曼不动杆菌的最低抑菌浓度(MIC)、最低杀菌浓度(MBC);结合抑菌效果(MIC)测定其抑菌前后碱性磷酸酶(ALP)的水平改变,检测其对细菌细胞通透性的影响。结果乌梅、五味子、薄荷、赤芍、黄连、艾叶、连翘、黄芩颗粒剂悬液对泛耐药鲍曼不动杆菌具有不同程度的抑菌作用,其MIC依次为:3.2、3.2、51.2、51.2、51.2、51.2、51.2、204.8 mg/ml,MBC依次为:6.4、6.4、102.4、102.4、102.4、102.4、102.4、409.6mg/ml。除赤芍外,其余中药颗粒剂ALP抑菌前后差异有统计学意义(均P<0.05),乌梅和五味子作用后更明显。结论8种中药颗粒剂对泛耐药鲍曼不动杆菌有不同程度的体外抑菌作用,其抑菌作用可能是破坏细胞壁、改变细菌的细胞膜的通透性而实现的。Objective To investigate the antibacterial mechanism of 8 kinds of Chinese medicine granules,including Fructus mume,against acinetobacter baumannii in vitro.Methods The minimum inhibitory concentration(MIC)and minimum bactericidal concentration(MBC)of the suspensions of Fructus mume,Fructus Schisandrae schisandrae,Mint,Red peony Root,Rhizoma coptidis,Artemisia argyi,Forsythiae forsythiae,and Scutellaria Granules against pan-drug resistance Acinetobacter baumannii were determined by the test tube dilution method.The changes of alkaline phosphatase(ALP)level before and after inhibition were measured.According to the inhibitory effect(MIC),and the effects on bacterial cell permeability were detected.Results The suspensions of fructus mume,Fructus Schisandrae schisandrae,mint,red peony root,Rhizoma cordyceps sinensis,Artemisia argyi leaf,Forsythiae forsythiae,and Scutellaria Granules had antibacterial effects on acinetobacter baumannii to different degrees.Their MIC's were as follow:3.2,3.2,51.2,51.2,51.2,51.2,51.2,51.2,51.2,51.2,and 204.8 mg/ml,and their MBC's 6.4,6.4,102.4,102.4,102.4,102.4,102.4,102.4,102.4 and 409.6 mg/ml,respectively.Except for red peony root,the differences of ALP in other Chinese medicine granules between before and after antibacterial were statistically significant(all P<0.05),and the differences were more obvious after the effect of black plum and Schisandra chinensis.Conclusion The antibacterial effect of 8 kinds of Traditional Chinese medicine granules on pan-drug resistant Acinetobacter baumannii in vitro may be realized by destroying cell wall and changing the permeability of cell membrane.
