摘  要:目的:探讨不同缺氧时间对单侧颈总动脉结扎新生大鼠脑损伤的影响。方法:7 d龄SD大鼠48只,随机分为假手术组(Sham)和缺氧缺血组(HI)。HI组双线结扎左侧颈总动脉并剪断。根据缺氧时间HI组又分为缺氧1.5 h组(HI-S)、缺氧2 h组(HI-M)和缺氧2.5 h组(HI-L)。术后恢复1 h后置于38℃密闭低氧箱中,并通入8%O2+92%N2混合气体。2,3,4-氯化三苯基四氮唑(TTC)染色评价脑梗死体积,激光散斑血流成像系统监测局部脑血流(r CBF)变化,苏木素-伊红(HE)染色观察大脑的病理形态学改变。结果:TTC染色结果显示HI-L组脑梗死体积大小基本一致;局部脑血流监测结果显示HI-M组和HI-L组损伤侧脑血流显著减少(P <0.05);HE染色结果显示HI-L组损伤侧大脑皮层及海马组织结构明显松散、细胞排列紊乱、细胞核固缩并裂解,出现大量空泡现象。结论:本研究中缺氧2.5 h时制备的模型脑损伤程度比较稳定,且成功率高,为进一步研究缺氧缺血性脑损伤的机制提供了可靠的动物模型。Objective:To explore the effects of duration of hypoxia on brain injury of neonatal rats with unilateral common carotid artery ligation.Methods:Forty-eight 7d old SD rats were randomly divided into sham group,and hypoxia-ischemia group.In hypoxia-ischemia group the rats were ligated with two wires on the left carotid artery which was cut in the middle.Based on the time for hypoxia,HI group was sub-divided into group HI-S(1.5 h for hypoxia),group HI-M(2 h for hypoxia),and group HI-L(2.5 h for hypoxia).After recovery,the rats were placed in Automatic Animal Hypoxia Laboratory(8%oxygen balanced with 92%nitrogen)at 38℃.Rats in sham group were treated with anesthesia and with their left carotid artery exposed,without the ligation of artery and hypoxia.The rats in both groups were sacrificed 24 h after the modelling and the brain tissue was removed for morphological observation.The volume of brain infarction was evaluated by TTC staining;Regional cortical blood flow(rCBF)was monitored by laser speckle contrast imaging(LSCI);The pathomorphological changes of the brain were observed by the use of HE staining.Results:TTC staining showed that the volume of cerebral infarction stays the same in group HI-L;rCBF monitoring showed that blood flow in the infarcted part of the brain was significantly reduced in group HI-M,and HI-L(P<0.05);HE staining showed the structure of cerebral cortex and hippocampus on the injured part of the brain is loose,accompanied with irregular neuron arrangement,the nucleus pycnosis,cracking,karyopyknosis,and cellular vacuolization in the rats of group HI-L significantly.The rats in sham group showed no significant changes.Conclusion:In this study,the degree of brain damage of the model prepared at 2.5 h of hypoxia is relatively stable,and the success rate is high,which provides a reliable animal model for further research on the mechanism of hypoxic-ischemic brain damage.
