摘  要:目的研究和分析不同病理分期人卵巢癌移植瘤组织趋化因子CXCL12表达量和携CXCL12靶向超声造影成像参数的关系。方法50只裸鼠用于制备人卵巢癌移植瘤模型,造模成功后每间隔10 d,随机取10只行非靶向及靶向超声造影成像,用造影分析软件获得造影参数;随后处死裸鼠行病理检查和RT-PCR技术得到各组卵巢癌组织CXCL12因子mRNA相对表达量,根据病理Ⅰ期、Ⅱ期、Ⅲ期分3组;分析不同病理分期靶向与非靶向超声造影时间强度曲线参数与卵巢癌组织中CXCL12因子RNA表达量的相关性。结果明确病理分期的37例移植瘤纳入本研究;时间强度曲线参数峰值强度(PI)和达峰时间(TTP)在CXCL12靶向微泡组于Ⅰ、Ⅱ、Ⅲ病理分期差异有统计学意义(P<0.05);与普通造影剂比较,靶向超声造影PI值在Ⅰ、Ⅱ期卵巢癌移植瘤组中较高,差异有统计学意义(P<0.05),靶向超声造影TTP值在Ⅰ期卵巢癌移植瘤组较小,差异有统计学意义(P<0.05);CXCL12靶向超声微泡PI与Ⅱ、Ⅲ期癌组织中CXCL12表达量呈正相关(P<0.05),参数TTP与Ⅰ、Ⅲ期癌组织中CXCL12表达量呈正相关(P<0.05),其余参数在各病理分期与瘤组织CXCL12 mRNA表达量无相关性(P>0.05)。结论携CXCL12抗体靶向微泡超声造影参数PI和TTP在人卵巢癌移植瘤不同病理分期存在差异,且与人卵巢癌移植瘤中CXCL12 mRNA相对表达量具有相关性。Objective To investigate the correlation between CXCL12 expression in human ovarian cancer transplanted tumor tissues with different pathological stages and parameters of CXCL12-targeted contrast-enhanced ultrasound.Methods A total of 50 nude mice were used to prepare human ovarian cancer xenograft model.After successful modeling,10 were randomly selected for untargeted and targeted contrast-enhanced ultrasound imaging every 10 days.Subsequently,nude mice were sacrificed for pathological examination and RT-PCR test on the relative expression of CXCL12 mRNA in ovarian cancer tissues of each group.The correlation between the time intensity curve parameters of targeted and untargeted contrast-enhanced ultrasound in different pathological stages and the expression of CXCL12 factor RNA in ovarian cancer tissue were analyzed.Results Thirty-seven cases of transplanted tumors with definite pathological stage were included in this study.The differences of time intensity curve parameters PI and TTP in the cxcl12-targeted microvesicle group in the pathological stages of Ⅰ,Ⅱ and Ⅲ were statistically significant(P<0.05).CXCL12 targeted ultrasound microbubble PI was positively correlated with the expression of CXCL12 in the stage Ⅱ and Ⅲ cancer tissues(P<0.05),and the parameter TTP was positively correlated with the expression of CXCL12 in the stage Ⅰ and Ⅲ cancer tissues(P<0.05).Conclusion The parameters PI and TTP of targeted microbubble contrastenhanced ultrasound with CXCL12 antibody are different at different pathological stages of human ovarian cancer transplanted tumors,and are related to the relative expression of CXCL12 mRNA in human ovarian cancer transplanted tumors.
