摘  要:探讨本课题组自主发现的人类内源多肽MIAC(Micropeptide inhibiting actin cytoskeleton)对头颈癌细胞CAL27增殖、迁移和凋亡的影响。采用LC-MS/MS检测CAL27细胞内源表达MIAC的情况;MTT法检测多肽MIAC对CAL27细胞增殖的影响;采用Transwell小室检测多肽MIAC对CAL27细胞迁移的影响;通过流式细胞仪检测多肽MIAC对CAL27细胞凋亡的影响。CAL27细胞能够内源性表达MIAC多肽,与对照组相比,多肽MIAC可以显著抑制CAL27细胞的增殖和迁移,促进CAL27细胞凋亡。由此说明多肽MIAC可以作为潜在的药物,通过抑制头颈癌细胞增殖、迁移,促进细胞凋亡发挥抗肿瘤作用。To explore the effect of human endogenous peptides MIAC(micropeptide inhibiting actin cytoskeleton),which was independently discovered by our group,on the cell proliferation,migration and apoptosis of head and neck cancer cells,the endogenous expression of MIAC in CAL27 cells was detected by LC-MS/MS,MTT assay was used to examine the effect of MIAC on CAL27 cellproliferation,transwell assay was performed to assess the CAL27 cellmigration with MIAC treatment,the effects of MIAC on CAL27 cell apoptosis was observed by Flow cytometry.The author’s data showed that CAL27 cells could endogenously express MIAC micropeptide,compared with the control group,the peptide MIAC could significantly inhibit the proliferation and migration of CAL27 cells,and MIAC peptide markedly promoted CAL27 cell apoptosis.The author’s results indicate that the micropeptide MIAC could be used as a potential drug to play an anti-tumor role by inhibiting the cellproliferation,migration and tumorigenesis of head and neck cancer.
