摘 要:为了研究槲皮苷通过在缺氧环境下对PC12细胞的保护作用及其机制,通过培养基中添加Co Cl2模拟慢性缺氧环境,通过MTT实验和Hoechst 33258染色实验研究在缺氧环境下槲皮苷对PC12细胞活性和细胞损伤的影响,通过流式细胞实验研究槲皮苷对PC12细胞凋亡的影响,通过蛋白免疫印迹Akt信号通路相关蛋白的表达情况。结果证明槲皮苷可显著增加PC12细胞在缺氧环境下的细胞活性,降低乳酸脱氢酶(Lactate dehydrogenase,LDH)含量,逆转缺氧环境下细胞细胞核固缩等细胞损伤。流式细胞实验结果显示,槲皮苷可显著降低PC12细胞凋亡率。蛋白免疫印迹实验结果显示,槲皮苷可逆转缺氧环境下PC12细胞中p-Akt表达下降和Cleaved caspase-3表达升高,并升高Bcl-2/Bax比值。槲皮苷通过激活Akt信号通路提高缺氧环境下神经细胞活性并降低凋亡水平。To study the protective effect of quercetin on PC12 cells under hypoxia and its specific mechanism,the effects of quercetin on PC12 cell viability and cell damage were studied by MTT and Hoechst 33258 staining experiments.The effect of quercetin on apoptosis of PC12 cells was studied by flow cytometry.The expression of Akt signaling pathway-related proteins was detected by Western blotting.Quercetin could significantly increase the cell activity of PC12 cells under hypoxic environment,reduced the content of lactate dehydrogenase(LDH),and reversed cell damage such as nuclear pyknosis under hypoxic environment.Flow cytometry showed that quercitrin could significantly reduce the apoptotic rate of PC12 cells.Western blotting showed that quercitrin could reverse the decrease of p-Akt expression and the increase of cleaved Caspase-3 expression in PC12 cells under hypoxia,and increase the Bcl-2/Bax ratio.Quercetin enhanced neuronal activity and reduced apoptotic level in hypoxic environment by activating Akt signaling pathway.
