摘 要:对广东省某规模化肉鸽养殖场暴发的疑似鸽痘进行了临床观察、病毒分离鉴定、动物回归试验和4b核心蛋白基因序列分析。成功分离得到1株皮肤型鸽痘病毒KP3-PPV。将KP3-PPV接种7日龄乳鸽后发现乳鸽从接种后第5天在翼膜接种部位开始出现黄色增生性结节,在接种后第18天在鼻翼、嘴角周围开始出现黄色增生性结节。扩增KP3-PPV的4b基因序列并进行比较分析,结果显示该分离株与南非FeP2株鸽痘病毒的同源性达100%,与2株企鹅痘病毒和火烈鸟痘病毒的同源性分别为99.36%、99.11%和98.05%,与鸡痘病毒的同源性为92.6%~93.09%,与其他禽痘病毒的同源性仅为76.03%~77.78%。本研究为鸽痘病理学研究、禽痘病毒间种属鉴定、鸽痘致弱疫苗开发提供了重要参考依据。The present study was performed for clinical observations,virus isolation and identification,animal regression experiment and sequence analysis of 4b core protein gene of a suspected pigeon pox from an outbreak in a large-scale meat pigeon farm of Guangdong Province.The KP3-PPV,a strain of skin-type pigeonpox virus(PPV)was successfully isolated.It was found that the yellow hyperplastic nodule began to appear in the pterygoid inoculation site from the 5th day after inoculating the KP3-PPV to 7-day-old pigeons,and around the nose wing and mouth corner from the 18th day after artificial inoculation.The 4b gene sequence of KP3-PPV was compared and analyzed,and the results showed that the KP3-PPV had 100%homology to South African FeP2 strain of PPV,99.36%and 99.11%homology to two strains of penguinpox virus,98.05%homology to flamingopox virus,92.6%to 93.09%homology to fowlpox viruses,76.03%to 77.78%homology to pox viruses of other birds.This study provides an important reference for the pathological research of pigeon pox,the identification of avipoxvirus species,and the development of pigeon pox attenuated vaccines.
