摘 要:旨在研究杨树花多糖(PFP)对大肠杆菌感染河田鸡的防治效果。将225只1日龄河田鸡随机分为9组,分别为空白组、模型对照组、阳性对照组与预防组(高、中、低)、治疗组(高、中、低),每组25只,连续用药6 d后,观察7 d。通过Western-blot检测TLR4、Myd88及NF-κB蛋白水平,通过qPCR检测IL-6、TNF-α的m RNA表达水平,初步探讨其作用机制。观察发现,杨树花多糖防治组鸡的精神状态和模型组相比显著改善,组织器官的症状及病理变化显著减轻。Western-blot结果显示,杨树花多糖防治组TLR4/NF-κB信号通路相关蛋白表达量下调明显,且呈现出剂量依赖型。qPCR结果显示,杨树花多糖防治组IL-6、TNF-α的表达量显著低于模型对照组(P<0.05),其中预防高剂量组IL-6 mRNA表达量与恩诺沙星组差异不显著(P>0.05),但TNF-α mRNA表达量显著低于恩诺沙星组(P<0.05)。结果说明,杨树花多糖能有效清除大肠杆菌的毒力因子,对河田鸡大肠杆菌病有较好的防治效果。The purpose of this study was to investigate the effect of poplar flower polysaccharide(PFP)on the Escherichia coli infection in Hetian chicken.A total of 225 Hetian chickens of 1-day-old were randomly divided into 9 groups(each group containing 25 chickens),such as blank group,model control group,positive group,prevention groups with PFP at high,medium and low dosage,treatment groups with PFP at high,medium and low dosage.After continuous treatment for 6 days,the 9 groups were observed for 7 days.In order to explore the mechanism of this experiment,the levels of TLR4/NF-κB-related protein expression and genes expression were detected by Western-blot and qPCR,respectively.The mental state,symptoms and pathological tissues in the prevention groups and treatment groups were significantly reduced than that in the model control group.The result of Western-blot showed that the expression of proteins related to the TLR4/NF-κB signaling pathway in the PFP prevention and treatment groups was significantly down-regulated in a dose-dependent manner.The result of qPCR showed that the expression levels of IL-6 and TNF-αmRNA in the PFP prevention and treatment groups were significantly lower than the model control group(P<0.05),and the expression level of IL-6 mRNA in the high-dose prevention group was not significantly different from that of the enrofloxacin group(P>0.05),but the expression level of TNF-αmRNA was significantly lower than that of the enrofloxacin group(P<0.05).These results showed that the PFP could significantly scavenge the virulence factors of E.coli,which have a good effect on the prevention and treatment of Hetian chicken induced by colibacillosis.
