细丽外担菌侵染对油茶叶生理生化变化的影响

摘  要:【目的】油茶为世界四大木本油料植物之一,是我国特有的油料树种。为探究油茶茶饼病与生理生化指标的响应关系。【方法】以受细丽外担菌侵染的油茶叶为材料,通过可见光光度计测定不同感病程度的油茶叶片中叶绿素(a+b)含量、丙二醛(MDA)含量、游离脯氨酸含量、可溶性糖含量、超氧化物歧化酶活性(SOD)、过氧化物酶活性(POD)、苯丙氨酸解氨酶(PAL)等生理生化指标。【结果】1)叶绿素和类胡萝卜素的含量呈下降趋势,且实验组明显低于对照组,呈极显著差异(P <0.01)。在病害最严重的第IV级,两者的含量分别为对照组的7.36%和2.38%;2)可溶性糖含量、脯氨酸含量、丙二醛含量在Ⅱ级达到峰值,分别是对照组的4.15倍、3.13倍和3.82倍,而后随病害加重三者含量呈下降趋势;与对照组含量呈显著变化(P <0.01),但这些指标均存在先上升至峰值而后急剧下降的趋势。3)茶饼病叶片中苯丙氨酸解氨酶(PAL)的活性变化呈显著差异(P <0.05),受病程的不断加重,其活性先上升至峰值而后急剧下降。4)超氧化物歧化酶活性和过氧化物酶活性呈下降趋势,不同病程的油茶叶中氧化酶活性存在显著差异(P <0.05),且始终远低于对照组(P <0.01)。单因素方差分析表明,在8个生理生化参数间,除可溶性糖的对照组与IV级病程组不存在显著性差异外(P <0.05),其余参数间,对照组与实验组均存在极显著差异(P <0.01)。【结论】细丽外担菌胁迫能够引起油茶叶的生理生化呈现不同程度的变化,为深入研究油茶与细丽外担菌的互作机理提供理论基础。【Objective】Camellia oleifera is a typical Chinese oil tree, which is one of the four largest woody oil plants in the world. To explore the resistance relationship between physiological & biochemical indices and oil-tea blister blight C. oleifera leaves induced by Exobasidium gracile.【Method】To explore the resistance relationship between physiological & biochemical indices and oil-tea blister blight C. oleifera leaves induced by E. gracile. The research investigated C. oleifera leaves infected by E. gracile at four stages labeled I-IV, where the physiological & biochemical indice such as chlorophyll(a+b), malondialdehyde(MDA), free proline, soluble sugar, superoxide dismutase(SOD), peroxidase(POD), phenylalanine ammonia lyase(PAL)are tested using visible light spectra. 【Result】1) Contents of chlorophyll and carotinoid kept descending, significantly lower than the control(P < 0.01), with 7.36% and 2.38% at Stage IV;2) The contents of soluble sugar, free praline and melondialdehyde(MDA), increased till the peak at stage Ⅱ, which were 4.15 times, 3.13 times and 3.82 times of the control group, respectively, and fell accordingly at other stages(P < 0.01), however, soluble sugar, proline content, MDA, activity all showed a tendency of rising to peak first and then decreasing sharply;3) The activity of phenylalanine ammonialyase(PAL) in the leaves infected by E. gracile showed a significant difference(P < 0.01) at four stages. With the aggravation of the disease course, its activity first increased to the peak and then decreased sharply. 4) The activities of superoxide dismutase(SOD) and peroxidase(POD) were on the decline and the oxidase activity at the four stages showed significant difference, lower than the control. It was found that, among the eight physiological parameter, except for soluble sugar, there was no significant difference between the control group and the group IV course(P < 0.05). However, there was a significant difference between the control group and the experimental group(P < 0.01).�
