摘  要:【目的】通过筛选花榈木高频率体胚发生的基因型和采种时期,为花榈木体胚发生的研究提供材料基础。【方法】花榈木7个基因型的成熟胚和5个采种时期的未成熟胚在B5培养基中添加6-BA(0.2 mg/L)+2,4-D(2.0 mg/L)+蔗糖(30 g/L)+结冷胶(2.5 g/L)+谷氨酰胺(0.5 g/L)进行胚性愈伤组织的诱导,其胚性愈伤组织在B5培养基中添加KT(0.5 mg/L)+2,4-D(1.0 mg/L)进行体胚诱导。【结果】1)在7个基因型中,基因型GZGL胚性愈伤诱导率最高(41.07%),基因型GZPT体胚诱导率最高(18.4%);胚性愈伤可溶性蛋白、可溶性糖和淀粉含量分别在基因型GZPT(7.21 mg/g)、AHQM(27.7 mg/g)和ZJLQ(18.39 mg/g)中最高。2)在不同采种时期中,胚性愈伤诱导率(37.49%)和体胚诱导率(14.2%)有一个窗口期,均以T4最高;胚性愈伤可溶性蛋白、可溶性糖和淀粉含量分别在T2(5.89 mg/g)、T1(18.39 mg/g)和T1(11.09 mg/g)中最高。3)相关性分析表明,体胚诱导率与基因型和采种时期具有显著相关性。4)利用细胞组织学技术初步了解到花榈木胚性愈伤细胞起源为内起源和外起源。【结论】不同基因型和采种时期对胚性愈伤诱导率、体胚诱导率及碳氮的积累具有较大的影响。其中,体胚诱导率以基因型GZPT最高;而在不同采种时期中,T4体胚诱导率最高。【Objective】High frequency somatic embryogenesis(SE)were screened by different genotypes and seed collecting stages of Ormosia henryi prain for providing an important basis for somatic embryogenesis induction.【Method】Mature embryos of seven genotypes and immature embryos of five seed collecting stages of Ormosia henryi prain were used as explants,Embryogenic Callus(EC)were induced in B5 medium supplemented with 0.2 mg/L 6-BA,2.0 mg/L 2,4-D,30 g/L sucrose,2.5 g/L gelatin,0.5 g/L glutamine,which induced somatic embryo in B5 medium supplemented with 0.5 mg/L KT,1.0 mg/L2,4-D.【Result】1)In seven genotypes,ECinduction rate and SE induction rate was highest in genotype GZGL(41.07%)and GZPT(18.4%)respectively,and the highest content of soluble protein,soluble sugar and starch in EC was the genotypes GZPT(7.21 mg/g),AHQM(27.7 mg/g)and ZJLQ(18.39 mg/g),respectively.2)At different seed collecting stages,there was a window period for the highest EC induction rate(37.49%)and SE induction rate(14.2%)in T4,and the highest content of soluble protein,soluble sugar and starch in EC was T2(5.89 mg/g),T1(18.39 mg/g)and T1(11.09 mg/g),respectively.3)The correlation analysis showed that SE induction rate had a significant correlation with genotype and seed collection period.4)The origin of EC cells of O.henryi was initially understood to be internal origin and external origin by histological techniques.【Conclusion】There were a significant effects on EC induction rate,SE induction rate,carbon accumulation and nitrogen accumulation in different genotypes and seed collecting stages.Among them,SE induction rate was the highest in genotype GZPT,while T4 was the highest in different seed collection periods.
