摘  要:目的探讨黄芪水蛭提取物小肠吸收液对小鼠脑微血管内皮细胞(Bend.3)正常细胞及连二亚硫酸钠所致缺氧复氧损伤后Bend.3细胞炎症因子动态变化的影响。方法实验首先采用肠囊外翻法制备受试小肠吸收液;其次,采用CCK-8法,检测不同浓度的小肠吸收液对Bend.3细胞存活率的影响;采用酶联免疫吸附法,检测小肠吸收液对细胞缺氧复氧损伤后细胞炎症因子TNF-α、IL-1β、IL-6、T-cell的动态表达量的影响。结果各药物干预组,细胞状态优于模型组。药物组小肠吸收液可提高损伤细胞的增殖率最高达到54.02%。各药物干预组,TNF-α含量在4~24 h均维持较低表达,与模型对照组比较,差异均有统计学意义(P <0.05,P <0.01);IL-6含量在8~24 h维持不同程度的低表达水平(P <0.05,P <0.01);T-cell含量在8 h呈现低表达水平(P <0.05,P <0.01);中、小剂量组IL-1β的含量,在24 h与模型组比较差异有统计学意义(P <0.05)。结论在本次试验条件下,黄芪水蛭小肠吸收液对正常Bend.3细胞无影响;对模型细胞炎症因子的表达量有一定的降低作用,表明黄芪水蛭小肠吸收液对Bend.3细胞缺氧复氧损伤后的炎症反应有改善作用。Objective To investigate the effects of extracts of Huangqi( Astragalus) and Shuizhi( Leech) on the dynamic changes of inflammatory factors in normal brain microvascular endothelial cells( Bend. 3) and bend. 3 cells after hypoxia-reoxygenation injury induced by sodium dithionite in mice. Methods Firstly,the small intestine absorbent solution was prepared by inversion of intestinal sac. Secondly,CCK-8 method was used to detect the effect of different concentrations of small intestinal absorption solution on the survival rate of Bend. 3 cells. Enzyme-linked immunosorbent assay( ELISA) was used to detect the effects of intestinal absorption solution on the dynamic expression of inflammatory cytokines IL-1β,IL-6,TNF-α and T-cell after hypoxia-reoxygenation injury. Results The cell status of each drug intervention group was better than that of the model group. The absorption of small intestine in the drug group can increase the proliferation rate of injured cells up to 54. 02%. The levels of TNF-α in each drug intervention group maintained a low expression from 4 to 24 hours. Compared with those of the model control group,the differences were statistically significant( P < 0. 05,P < 0. 01). The levels of IL-6 in each drug intervention group maintained low expression levels at different levels from 8 to 24 hours( P < 0. 05,P < 0. 01). T-cell content showed low expression level at 8 h( P < 0. 05,P < 0. 01). The content of IL-1β in the middle and low dose groups was statistically different from that in the model group at 24 h( P < 0. 05). Conclusion The results showed that Huangqi( Astragalus) and Shuizhi( Leech) intestinal absorption solution had no effect on normal Bend. 3 cells,and had a certain effect on the expressions of inflammatory factors in model cells. These results suggest that Huangqi( Astragalus) and Shuizhi( Leech) intestinal absorption solution can improve the inflammatory response of Bend. 3 cells after hypoxia-reoxygenation injury.
