摘 要:目的研究蒙药紫花高乌头不同溶剂萃取部分对慢性支气管炎(CB)大鼠的治疗作用,并初步阐释其治疗作用机制。方法采用系统溶剂法对蒙药紫花高乌头80%乙醇提取物进行萃取,分别得到石油醚、三氯甲烷、乙酸乙酯、正丁醇及乙醇萃取部分;采用脂多糖(LPS)气管滴入(200μg·200μL-1)联合SO2吸入法建立CB大鼠模型;蒙药紫花高乌头石油醚、三氯甲烷、乙酸乙酯、正丁醇及乙醇萃取部分分别口服灌胃给药,并用ELSA法测定大鼠肺组织匀浆中肿瘤坏死因子-α(TNF-α),白细胞介素(IL-4、IL-10)含量变化。结果建立了CB大鼠模型,得到了蒙药紫花高乌头不同溶剂萃取部分;与模型组相比,蒙药紫花高乌头三氯甲烷、乙酸乙酯及正丁醇萃取部分可以改善CB大鼠的反应迟钝、毛色枯槁、进食减少及体质量不增等情况;与空白组相比,模型组大鼠肺组织匀浆中TNF-α含量显著升高(P <0.05或P<0.01),IL-4、IL-10含量显著降低(P <0.05或P <0.01);与模型组相比,蒙药紫花高乌头三氯甲烷、乙酸乙酯及正丁醇萃取部分高低剂量组大鼠肺组织匀浆中TNF-α含量显著降低(P <0.05或P <0.01),IL-4、IL-10含量显著增高(P <0.05或P <0.01)与阳性组相比,蒙药紫花高乌头三氯甲烷、乙酸乙酯及正丁醇萃取部分高低剂量组大鼠肺组织匀浆中TNF-α、IL-4及IL-10含量差异无统计学意义(P> 0.05)。结论蒙药紫花高乌头对CB大鼠治疗作用的活性部位为三氯甲烷、乙酸乙酯及正丁醇萃取部分,其中三氯甲烷萃取部分活性最强,石油醚与乙醇萃取部分活性最弱。蒙药紫花高乌头可以抑制炎症因子的释放,增强抗炎细胞因子活性,调节细胞因子间失衡状态,从而对CB大鼠产生治疗作用。Objective To evaluate the effect ofdifferent extraction parts of Aconitum excelsum Reichb on chronic bronchitis( CB) rats and their mechanism. Methods Systematic solvent method was used to extract the 80% ethanol extracts of Aconitum excelsum Reichb,and five different extraction fractions( petroleum ether,chloroform,ethyl acetate,n-butanol and alcohol) were obtained. The rat model of CB was established with lipopolysaccharide( LPS,200 μg·200 μL-1) by tracheal injection. At the same time,rats were given sulfur dioxide( SO2) inhalation. Petroleum ether,chloroform,ethyl acetate,n-butanol and alcohol from Aconitum excelsum Reichb were administrated,and the levels of tumor necrosis factor( TNF-α),interleukin-4( IL-4) and interleukin-10( IL-10) were determined by ELISA method. Results The rat model of CB was established,different extraction fractions of Aconitum excelsum Reichb were obtained. Compared with those of the model group,the administration of chloroform,ethyl acetate and n-butanol extract could improve the tiredness and weakness of CB rats,reduced food intake and body weight without increasing status. Compared with those of the blank group,the increase of TNF-α,the reduce of IL-4 and IL-10 in the lung tissue of rats with CB in the homogenate had statistical significance( P < 0. 05 or P < 0. 01). The high and low dose of chloroform,ethyl acetate and n-butanol extract could significantly inhibit the increase of TNF-α,at the same time,could significantly inhibit the reduce of IL-4 and IL-10 in the lung tissue of rats with CB in the homogenate. Compared with those of the model group,there was a statistical significance( P < 0. 05 or P < 0. 01),but compared with those of the positive group there was no significant difference( P > 0. 05). Conclusion The chloroform,ethyl acetate and n-butanol extracts of Aconitum excelsum Reichb have the better treatment effect on CB.
