摘  要:反硝化细菌常用于污染水体的治理与修复。为探究好氧反硝化细菌的加入对水体中原位细菌群落的影响,以污染湖泊水环境为研究对象,在湖水中设置3根试验管并分别添加0g、2g、20g地衣芽孢杆菌菌粉,连续取4次水样后,利用基因定量和高通量测序技术对湖泊水体细菌群落中的好氧反硝化基因拷贝数和细菌群落组成及代谢功能进行研究。结果表明:好氧反硝化基因napA的拷贝数变化显示,好氧反硝化细菌在湖泊浅层水体中的存活时间为4d左右;添加外源反硝化细菌会降低湖泊水体中原位细菌群落多样性,改变细菌群落结构,其中Gammaproteobacteria的相对丰度与外源反硝化细菌投加量呈正相关,与时间呈负相关,而Betaproteobacteria、Alphaproteobacteria、Acidimicrobiia、Actinobacteria的相对丰度均与外源反硝化细菌投加量呈负相关;投加外源反硝化细菌短时间内可以增强湖泊水体中细菌群落的反硝化能力,但随着时间的延长,原位微生物会恢复其竞争优势。Denitrifying bacteria is applied widely in polluted water bioremediation and treatment.In order to study the influence of aerobic denitrifying bacteria inoculation on original bacterial communities in water,this paper sets up three experimental tubes inserted in a lake,with 0g,2g,and 20g Bacillus licheniformis powder added into,respectively.Taking the water samples four times,the paper adopts gene quantification and high-throughput sequencing technologies to study the copy numbers of denitrification genes,the composition and metabolic functions of the bacterial communities in water.The results show that the change of copy number of aerobic denitrification gene napAsuggests that inoculated denitrifying bacteria can survive no longer than 4days in shallow water.Inoculated denitrifying bacteria reduces the bacterial community’s diversity,and changes the community composition.The relative abundance of Gammaproteobacteria has a positive correlation with the powder inoculation volume,and a negative correlation with time,while the relative abundance of Betaproteobacteria,Alphaproteobacteria,Acidimicrobiia and Actinobacteria has negative correlation with powder inoculation volume.Inoculated denitrifying bacteria increases the denitrification ability of bacterial community in short time,but the original bacterial community’s competitive advantage will be restored in long term.
