摘  要:为探究甘油对湖羊精液常温保存效果的影响,在基础稀释液中添加0 (对照组)、2%、4%、6%和8%甘油,检测16℃保存过程中湖羊精子活率、活力、质膜完整率、顶体完整率和直线速率等运动性能指标。结果表明:与对照组相比,保存48~120 h时, 2%甘油组精子活率(P>0.05)、质膜完整率(P<0.05)和顶体完整率(P>0.05)提高;保存144~168 h时, 2%和4%甘油组精子活率(P>0.05)、质膜完整率(P<0.05)和顶体完整率(P>0.05)提高;保存192 h时,各甘油组精子活率(P<0.05)提高。随着甘油浓度的增加,直线速率、曲线速率、平均路径速率和侧摆幅度也表现为先升高后降低的趋势。在试验条件下,保存24 h内各甘油组均降低精液品质,保存48~120 h时2%甘油组精液品质提高,保存144~168 h时2%和4%甘油组精液品质提高,保存192 h时各甘油组精液品质均提高。In order to explore the effect of adding different concentrations of glycerol to the diluent at room temperature on the preservation effect of Hu ram semen, different concentrations of glycerol 0(Control), 2%, 4%, 6% and 8% were added to the basic diluent to detect the motility, viability, plasma membrane integrity rate, acrosomal integrity rate, motility parameters and linear rate of Hu ram sperm during preservation at 16 ℃. The results showed that compared with the control group, the sperm motility(P>0.05), plasma membrane integrity rate(P<0.05) and acrosome integrity rate(P>0.05) were increased in the 2% glycerol group storing for 48-120 h. Compared with the control group, sperm motility(P>0.05), plasma membrane integrity rate(P<0.05) and acrosome integrity rate(P>0.05) were increased in the 2% and 4% glycerol groups storing for 144-168 h. Compared with the control group, the sperm motility of each glycerol group was increased after preservation for 192 h(P<0.05). With the increase of glycerol concentration, the linear rate, curve rate, average path rate and swing amplitude were also increased at first and then decreased significantly in 6% and 8% glycerol groups. Therefore, under the condition of this experiment, the preservation effects of glycerol with different concentration and different preservation time on Hu ram semen were different with those under the condition of normal temperature. The semen qualities of all glycerol groups decreased within 24 h of preservation. Stored for 48-120 h, 2% glycerol group improved semen quality. Stored for 144-168 h, 2% and 4% glycerol group improved semen quality. After being preserved for 192 h, the semen qualities were improved in all glycerol groups.
