摘  要:为了探究大岩桐育种技术,本研究以大岩桐植物(Sinningia speciosa)花粉为试材,对其花粉萌发特性进行试验。利用离体萌发法探究了蔗糖、硼酸、GA3、硝酸钙等单因素及多因素组合对大岩桐花粉萌发的影响,并在此基础上探究了不同Ca2+、Mg2+浓度下花粉管的生长速度。结果表明,适合大岩桐花粉萌发的培养基为蔗糖100 g/L+硼酸10 mg/L,萌发率为75.67%;适合大岩桐花粉管生长的Ca2+浓度为250 mg/L,培养38 h后花粉管生长最长可达846.21μm;适合大岩桐花粉管生长的Mg2+浓度为400 mg/L,培养38 h后花粉管生长最长可达815.45μm。通过对大岩桐花粉萌发特性的探究,为大岩桐新品种培育提供理论依据。In order to explore the breeding technology of Sinningia speciosa, the pollen of Sinningia speciosa was used as material, pollen germination characteristics were tested. The effects of sugar, boricic acid and GA3, lime nitrate etc on pollen germination were studied by in vitro germination, and on this basis, the growth rate of pollen tube under different concentrations of Ca^2+, Mg^2+was discussed. The results showed that the suitable solid agar medium for pollen germination was sucrose 100 g/L+boric acid 10 mg/L, and the germination rate was 75.67%.The optimum concentration of Ca2+for pollen tube growth of Sinningia speciosa is 250 mg/L. The longest pollen tube growth was 846.21 μm after 38 hours of culture. The optimum concentration of Mg^2+for pollen tube growth of Sinningia speciosa is 400 mg/L. The longest pollen tube growth was 815.45 μm after 38 hours of culture.Through exploring the pollen germination characteristics of Sinningia speciosa, the theoretical basis for the cultivation of new varieties of Sinningia speciosa was provided.
