藏药五脉绿绒蒿多糖提取工艺实验研究

摘  要:以青海种植藏药五脉绿绒蒿为原料,采用苯酚-硫酸显色法和紫外-可见分光光度法测定了藏药五脉绿绒蒿多糖含量;用单因素实验法与响应面法确定溶剂萃取-超声波辅助提取多糖的优化工艺。结果显示影响多糖提取率因素顺序为:提取温度>液料比>提取时间,在溶剂萃取压力为2MPa、超声波功率为200W、提取温度为67℃、提取时间为32min、液料比为27∶1(mL/mg)的最优工艺条件下,重复实验3次,多糖的平均提取率为74.68%,相对标准偏差为0.12%,与理论预测值74.87%基本吻合,该优化工艺能显著提高多糖的提取率,提取时间短,操作简便、稳定。A Tibetan medicine,Meconopsis quintuplinenia Regel,planting in Qinghai Province was used as raw material for exploring optimal extraction process of the polysaccharides and content of the polysaccharide was determined by phenol-sulfuric acid colorimetric method.The ultrasonic assisted solvent extraction method was adopted to optimize the extraction process,which was analyzed by single factor test and response surface method.The results showed that the temperature,ratio of solvent to material and extraction time affected the yield of the polysaccharide in descending order.Under the optimum conditions,2MPa of extraction pressure,200W of ultrasonic power,67℃of extraction temperature,32min of extraction time and 27∶1(mL/mg)of ratio of solvent to material,repeat the experiment three times,the average yield of polysaccharide was 74.68%±0.12%,which was almost consistent with the theoretical prediction value of 74.87%.The optimized process significantly improved the yield of polysaccharide in short time,which was easy to operate and stable.
