复方苁蓉精水提液对帕金森病模型大鼠中脑GSK-3β表达的影响

摘　　要：目的研究复方苁蓉精水提液对PD模型大鼠行为学的影响及其对中脑黑质—纹状体组织中糖原合成酶激酶3β(glycogen synthase kinase 3β,GSK-3β)蛋白表达的影响。方法取SD大鼠50只,随机分为复方苁蓉精水提液低、中、高剂量组,模型组和正常组五组,采用颈背部皮下注射鱼藤酮葵花油乳化液制备PD大鼠模型,干预14天后,进行行为学观察、实验,提取中脑黑质,用免疫荧光法检测GSK-3β表达情况。结果(1)经复方苁蓉精水提液干预14天,中剂量和高剂量组大鼠GSK-3β的表达较模型组来说显著降低(P<0.05);(2)经复方苁蓉精水提液干预14天后,中剂量和高剂量组悬挂实验行为学评分增加(P<0.05)。结论复方苁蓉精水提液可能通过抑制GSK-3β的活性,以此来改善PD大鼠的行为障碍。Objective The research is to study the effect of Cistanchedeserticola extract on the behavior of PD rats and the expression of GSK-3βprotein in subst-antianigra striatum of midbrain.Methods 50 SD rats were randomly divided into fivegroups:low,medium and high dose of Cistanche extract,model group and norm-al group.Rotenone sunflower oil emulsion was injected subcutaneously into the back of neck to establish PD rat model.After 14 days of intervention,behavioral observation and experiment were conducted.The substantia nigra striatum of midbrain was extracted to detect the expression of GSK-3βby immunofluorescence.Results After 14 days of intervention,the expression of GSK-3βin model group was significantly higher than that in normal group,middle dose group and high-dose group(P<0.05).There was no statistically significant difference between the low-dose group and the model group(P>0.05).Conclusion Cistanchedeserticola extract can improve the behavior disorder of PD rats by inhibiting the activity of GSK-3β.
