摘  要:肿瘤坏死因子受体相关因子6(TNF receptor associated factor 6,TRAF6)对于介导Toll样受体(TLR)/白细胞介素1受体(IL-1R)信号传导以及随后的NF-κB和AP-1的激活至关重要。在本研究中,已经克隆并鉴定了池蝶蚌(Hyriopsis schlegelii)TRAF6(以下简称HsTRAF6),cDNA全长为3945 bp,ORF框1965 bp,编码654个氨基酸,该区域包含保守的特征结构域,包括1个RING结构域、2个TRAF型锌指结构域、1个典型的卷曲螺旋(coiled coil;TRAF-N)和MATH(TRAF-C)结构域。系统发育分析显示,池蝶蚌与其他贝类等软体动物聚在一支。此外,qRT-PCR分析显示HsTRAF6在组织中广泛分布,在鳃和肝胰腺中表达最丰富;脂多糖和嗜水气单胞菌免疫刺激后,分别在7个时间点(0,6,12,24,36,48,72 h)对10个组织中HsTRAF6的mRNA表达差异性进行分析,各组织间表达变化存在差异,但均有显著变化。TRAF6(TNF receptor associated factor 6)is crucial for mediating Toll-like receptor(TLR)/Interleukin-1 receptor(IL-1R)signaling and subsequent activation of NF-κB and AP-1.In this study,TRAF6(hereinafter referred to as HsTRAF6)of Hyriopsis schlegelii has been cloned and identified.The cDNA is 3945bp in length and 1965bp in ORF frame,encoding 654 amino acids.This region contains conservative characteristic domains,including a RING domain,two TRAF-type zinc finger domains,and a typical coiled coil(TRAF-N)and MATH(TRAF-C)domain.Phylogenetic analysis showed that Hyriopsis schlegelii and other mollusks gathered in one group.In addition,qRT-PCR analysis showed that HsTRAF6 was widely distributed in tissues and expressed most abundantly in gills and hepatopancreas.After immune stimulation with LPS or Aeromonas hydrophila,the mRNA expression differences of HsTRAF6 in 10 tissues were analyzed at 7 time points(0,6,12,24,36,48,72 h).The expression changes between the tissues were different,but there were significant changes.
