摘 要:目的调查云南省保山市隆阳区捕获的小兽中是否携带鼠疫噬菌体,并探讨其流行病学意义。方法 2017年9月在保山市隆阳区板桥、辛街、河图、潞江4个乡(镇)8个自然村采集小兽标本,以鼠疫疫苗株EV76为宿主菌,采用双层琼脂平板法分离鼠疫噬菌体,描述其形态特征并分析流行病学意义。结果共捕获9种290只小兽,其中黄胸鼠139只、灰麝鼩57只、短尾鼩41只、大臭鼩31只,其他22只;290份标本中分离鼠疫噬菌体12株,总分离率4.14%,其中分离率大臭鼩12.90%、褐家鼠10.00%、黄胸鼠4.32%和灰麝鼩1.75%,其他鼠种未分离出;在兰村、辛街村、水头村、董家湾、下庄双寨、汪宣村6个调查点分离出鼠疫噬菌体,知青农场和下东村未分离到鼠疫噬菌体;挑选其中2株有代表性的鼠疫噬菌体进行电镜观察,均为肌尾噬菌体。结论保山市隆阳区小兽中存在鼠疫噬菌体,大臭鼩有较高的分离率;分离出的噬菌体对深入了解该地区鼠疫流行有一定意义。Objective To investigate whether there was Yersinia pestis phage or not in small mammals captured in Longyang district of Baoshan city,and explore their epidemiological significance.Methods The small mammals were captured from 8 villages at 4 townships of Banqiao,Xinjie,Hetu and Lujiang of Longyang District in Baoshan city in September,2017.The Yersinia Pestis phage were isolated with double-layer agar plate method by taking the host bacteria of vaccine strain EV76.The morphological characteristics were described and epidemiological significance were analyzed.Results There were 290 small mammals of 9 species captured,including 139 of Rattus flavipectus,57 of Crocidura attenuatas,41 of Anourosorex squamipes,31 of Suncus murinus and 22 of others.Totally 12 strains of Yersinia pestis phage were isolated with total isolation rate of 4.14%(12/290),including 12.90%(4/31)in Suncus murinus,10.00%(1/10)in Rattus norvegicus,4.32%(6/139)in Rattus flavipectus,and 1.75%(1/57)in Crocidura attenuatas.The Yersinia pestis phage were isolated at 6 investigation sites,that is,Lancun,Xinjie,Shuitou,Dongjiawan,Xiazhuangshuangzhai and Wangxuan,and not isolated in educated youth farm and Xiadong.Two strains of representative plague phages were selected for electron microscope observation,and both phages were Myoviridae family.Conclusions There are Yersinia pestis phages in small mammals in Longyang district of Baoshan city,and the isolation rate is high in Suncus murinus.All isolated phages are of certain significance for understanding the epidemic of plague in this area further.
