摘 要:目的探讨针刺腧穴"降压方"对自发性高血压大鼠(SHR)心肌肥厚的影响及可能作用机制。方法选择雄性WKY大鼠10只作为空白组,另选SHR 30只随机分为模型组、最小化针刺组和降压方组,每组10只。降压方组大鼠针刺双侧"人迎""曲池""足三里"穴,每日1次,连续针刺6天,休息1天;最小化针刺组仅浅刺各穴,不予行针,各组大鼠均干预8周。于干预前及干预第2、4、6、8周末测量大鼠尾动脉收缩压;干预后采用心脏超声检测各组大鼠室间隔舒张末厚度(IVSS)、左室舒张末期内径(LVIDd)、左室后壁舒张期厚度(LVPWd),计算心脏重量指数(HWI),运用WGA染色法观察各组大鼠心肌组织结构,采用RT-qPCR法检测心肌组织心房利钠肽(ANP)mRNA、β-肌球蛋白重链(β-MHC)mRNA表达。结果在干预前及干预第2、4、6、8周末,模型组大鼠尾动脉收缩压均显著高于空白组(P<0.05);在第4、6、8周,降压方组大鼠收缩压明显低于模型组和最小化针刺组(P<0.05)。与空白组比较,模型组大鼠IVSS、LVPWd、HWI以及心肌组织ANP和β-MHC mRNA表达增加,心肌细胞直径与横截面积均增大,LVIDd降低(P<0.05);与模型组比较,降压方组大鼠上述各指标均改善(P<0.05);而最小化针刺组大鼠各指标与模型组比较差异无统计学意义(P>0.05)。结论针刺腧穴"降压方"可有效改善SHR血压和心肌肥厚状态,可能是通过抑制心肌组织ANP和β-MHC mRNA表达来实现。Objective To explore the effects of acupuncture at acupoint "Jiangya Formula(降压方)" on myocardial hypertrophy in spontaneously hypertensive rats(SHR) and its possible mechanism. Method A total of 10 male WKY rats were selected as the blank group, and 30 SHR rats were randomly divided into a model group, a minimization acupuncture group and a Jiangya Formula group, with 10 rats in each group. Rats in the Jiangya Formula group were given acupuncture on both sides of "Renying(ST 9)", "Quchi(LI11)" and "Zusanli(S36)" points, once a day, for 6 consecutive days, with 1 day rest;the minimization acupuncture group was only shallowly punctured each point, without manupulation, and rats in each group were intervened for 8 weeks. The systolic blood pressure of the tail artery was measured before the intervention and on the end of the 2 nd, 4 th, 6 th, and 8 th week;after the intervention, cardiac ultrasound was used to detect the diastolic thickness of the ventricular septum(IVSS), the left ventricular end diastolic diameter(LVIDd), diastolic thickness of the posterior wall of the left ventricle(LVPWd), and to calculate the heart weight index(HWI);the myocardial tissue structure of rats in each group was observed by WGA staining method, and the expression of atrial natriuretic peptide(ANP) mRNA and β-myosin heavy chain(β-MHC) mRNA in myocardial tissue were detected by RT-qPCR method. Results Before the intervention and at the end of the 2 th, 4 th, 6 th, 8 th week intervention, the systolic blood pressure of the tail artery in the model group was significantly higher than that of the blank group(P<0.05);at the 4 th, 6 th, 8 th week, the systolic blood pressure of rats in the Jiangya Formula group was significantly lower than that in the model group and the minimization acupuncture group(P<0.05). Compared with the blank group, the expression of IVSS, LVPWd, HWI, myocardial tissue ANP and β-MHC mRNA in the model group increased, the diameter and cross-sectional area of myocardial cells increased, and the LVIDd decreas
