稳消方对兔动脉粥样硬化模型腹主动脉IκBα与NF-κB蛋白表达的影响

摘　　要：目的观察稳消方对兔动脉粥样硬化模型腹主动脉组织和Kappa B抑制蛋白α(IκBα)与核转录因子κB(NF-κB)蛋白表达的影响,探究稳消方防治动脉粥样硬化的作用机制。方法将38只清洁级新西兰大白兔随机分为空白组(9只)和造模组(29只),分别给予标准饲料和高脂饲料喂养12周后,按照随机数字表法将造模组随机分为模型组(灌胃生理盐水10 mL/kg)、稳消方组(半夏383 mg/kg,牡丹皮、郁金、丹参、水蛭、地龙各575 mg/kg)和辛伐他汀组(0.77 mg/kg),各9只,灌胃治疗8周后取材,以免疫组化法(IHC)及免疫蛋白质印迹法(Western Bolt)观察NF-κB和IκBα蛋白表达。结果与模型组比较,苏木精-伊红(HE)染色显示,稳消方组和辛伐他汀组主动脉内膜相对光滑完整,未见明显增生,仅可见少量斑块,免疫组化法和Western Bolt检测结果均显示,与模型组比较,稳消方组IκBα明显增加,NF-κB表达明显减少,差异均有统计学意义(P<0.01)。结论稳消方可通过增加IκBα蛋白水平,抑制NF-κB活化,减少促炎因子的过表达,表明稳消方对稳定斑块、抑制动脉粥样硬化具有一定作用。Objective To observe the pathological changes of abdominal aorta and the expression levels of I-kappa B alpha(IκBα)and NF-kappa B(NF-κB)protein in rabbits with atherosclerosis,and to explore the mechanism of Wenxiao Decoction in preventing and treating atherosclerosis.Methods Thirty-eight clean-grade rabbits were randomly divided into blank group(9 rabbits)and model group(29 rabbits).After 12 weeks of feeding with standard feed and high-fat feed,the rabbits in model group were randomly divided into model control group(saline 10 mL/kg),Wenxiao Decoction group(Pinellia ternata 383 mg/kg,danpi,tulip,Salvia miltiorrhiza,leech,and earthworm 575 mg/kg),and simvastatin group(0.77 mg/kg).After 8 weeks of intragastric administration,the samples were taken for observation of NF-κB and IκBαprotein expression by immunohistochemical method(IHC)and Western Blotting(WB).Results Compared with model control group,Hematoxylin-Eosin(HE)staining showed that aortic intima of Wenxiao Decoction group and simvastatin group were relatively smooth and intact,no significant increase was observed,only a few plaques could be seen.IHC and WB test results showed that,compared with model group,Wenxiao Decoction group could significantly increase the expression of IκBαin Wenxiao Decoction group(P<0.01)and inhibit the expression of NF-κB(P<0.01).Conclusion Wenxiao Decoction can increase the level of IκBαprotein,inhibit the activation of NF-κB,and reduce the overexpression of pro-inflammatory genes,which indicates that Wenxiao Decoction has a certain role in stabilizing plaques and inhibiting atherosclerosis.
