摘  要:目的观察霉茶总黄酮对自发性高血压大鼠(SHR)肾脏的保护作用,并根据研究肾小管上皮细胞(NRK-52E)对其作用机制进行探讨。方法 (1)将50只雄性SHR随机分为模型组,复方罗布麻片组(50 mg·kg-1),霉茶总黄酮低、中、高剂量(90,180,360 mg·kg-1)组,每组10只。各组每天早晚各灌胃给药1次,容量均为10 mL·kg-1体质量,连续给药7周。末次给药后1 h处死动物,迅速剥离肾脏组织,计算大鼠肾脏指数;HE染色观察肾脏组织结构形态变化;测定肾脏组织TLR4、 IL-6、 TNF-αmRNA的表达;(2)AngⅡ刺激NRK-52E后,经霉茶总黄酮各剂量(0.04,0.2,1,5μg·ml-1)干预24h,收集细胞,测定各组TLR4,IL-6, TNF-α的mRNA的表达。结果霉茶总黄酮可改善SHR肾脏组织的炎性病变,降低SHR肾脏组织和NRK-52E细胞中TLR4,IL-6, TNF-α的mRNA的表达水平(P<0.05或P<0.01)。结论霉茶总黄酮对SHR肾脏有一定的保护作用,其机制可能与下调炎性因子TLR4,IL-6,TNF-α的表达有关。Objective To observe the protective effect of Meicha total flavonoids on the kidney of spontaneously hypertensive rats(SHR), and to explore its mechanism of action in combination with renal tubular epithelial cells(NRK-52 E).Methods 50 male SHRs were randomly divided into model group, compound apocynum tablets group(positive drug,50 mg·kg-1), Meicha total flavonoids low-dose,medium-dose and high-dose groups(90,180,360 mg·kg-1), 10 in each group.Each group was intragastrically administered once a day in the morning and afternoon, and the capacity was 10 mL·kg-1 body weight for 7 weeks.Renal tissue was quickly excised after the last administration, and calculate the rat renal index;the histomorphological changes of the kidney were observed by hematoxylin-eosin(HE) staining;mRNA expression of Toll-like receptor 4(TLR4), interleukin-6(IL-6) and tumor necrosis factor-α(TNF-α) in renal tissues was determined by Real-time PCR.Angiotensin II( Ang II) stimulated normal rat renal tubular epithelial cells(NRK-52 E), and was treated with various doses of Meicha total flavonoids(0.04, 0.2, 1, 5 μg·ml-1) for 24 h, and the cells were collected for Real-time PCR analysis of mRNA expression of TLR4, IL-6 and TNF-α in each group.Results Meicha total flavonoids can improve the inflammatory lesions of SHR kidney tissue and decrease the expression of TLR4, IL-6 and TNF-α mRNA in SHR kidney tissue and NRK-52 E cells(P<0.05 or P<0.01).Conclusion Meicha total flavonoids has a certain protective effect on SHR kidney, and its mechanism may be related to down-regulation of mRNA expression of inflammatory factors TLR4, IL-6 and TNF-α.
