镧在UV-B应力下对迭鞘石斛生理及药用成分的影响

摘  要:为考察镧对UV-B应力下的迭鞘石斛[Dendrobium aurantiacum Rchb.f.var.denneanum(Kerr)]生理及药用成分的影响,对UV-B辐射下的迭鞘石斛叶面喷施硝酸镧溶液,筛选出最适合迭鞘石斛生长的镧溶液浓度,并考察在此浓度作用下迭鞘石斛的生理、抗氧化酶活性及药用成分的变化.UV-B辐射按强度分为T1(1.8 mmol m-2 s-1)、T2(3.6 mmol m-2s-1)、T3(5.4 mmol m-2 s-1),辐射时间共12 d,每3 d检测一次.结果表明,与其他浓度的镧溶液相比,20 mg/L镧溶液能促进迭鞘石斛的光合色素含量显著增加,因此最适宜迭鞘石斛生长.当迭鞘石斛接受低剂量UV-B(1.8 mmol m-2 s-1)辐射时,处理9 d时,镧元素仍然能显著促进光合作用,但当辐射强度较大(5.4 mmol m-2s-1)时,镧元素能显著促进光合作用的时间则降低为6 d.当迭鞘石斛接受5.4 mmol m-2 s-1的UV-B辐射12 d时,同时喷施镧溶液能显著增加可溶性糖和可溶性蛋白的含量,且显著增强过氧化氢酶(CAT)的活性,有利于缓解辐射造成的氧化伤害.当迭鞘石斛接受3.6 mmol m-2s-1 UV-B辐射3d时,喷施镧溶液能显著促进叶片中超氧化物歧化酶(SOD)活性,增强SOD清除超氧阴离子的能力.当迭鞘石斛接受低剂量(1.8 mmol m-2 s-1)UV-B辐射6 d,同时喷施镧溶液时,会显著促进多糖和多酚的积累,提升迭鞘石斛药用价值.本研究表明镧元素能有效保护UV-B辐射下的迭鞘石斛,并增加迭鞘石斛的药用价值.(图10参25)The study aimed to investigate the effects of lanthanum on the physiological and medicinal components of Dendrobium aurantiacum Rchb. f. var. denneanum(Kerr) under UV-B stress. In this study, a lanthanum nitrate solution was sprayed on the leaves of D. denneanum under UV-B radiation to find the most suitable concentration of lanthanum solution for the growth of D. denneanum. At the same time, the changes in some physiological indexes, antioxidant enzymes, and active components of D. denneanum were monitored. The experiment set three radiation intensities: low(1.8 mmol m-2 s-1), medium(3.6 mmol m-2 s-1), and high(5.4 mmol m-2 s-1). The radiation plus lanthanum treatment time was 12 days, and samples were tested every three days. Compared with other lanthanum concentrations, 20 mg/L lanthanum solution increases the total chlorophyll, and the quantity of carotenoids in the leaves significantly increased(P < 0.05). Therefore, this concentration is the most suitable for the growth of D. denneanum. It was found that, when D. denneanum was irradiated with UV-B for nine days(the radiation was 1.8 mmol m-2 s-1), the effect of spraying lanthanum solution on photosynthesis in the relevant group was reduced, and the synthesis of photosynthetic pigment was increased. However, when the radiation intensity was increased to 5.4 mmol m-2 s-1, the period during which lanthanum could significantly promote photosynthesis was reduced to six days. When D. denneanum was irradiated with UV-B for 12 days(the radiation was 5.4 mmol m-2 s-1), the content of soluble sugar and protein and the activity of catalase(CAT) in the group sprayed with lanthanum were significantly increased, all of which alleviate the oxidative damage caused by radiation. When D. denneanum was irradiated with 3.6 mmol m-2 s-1 UV-B for three days, spraying with lanthanum solution could significantly promote the activity of superoxide dismutase(SOD) and enhance the ability of SOD to scavenge superoxide anion.
