莲心碱对膀胱癌细胞T24增殖与自噬体形成的影响

摘　　要：目的研究莲心碱(Liensinine)抑制膀胱癌T24细胞增殖的作用并探讨其促进细胞自噬体形成的作用。方法以不同浓度的莲心碱作用于T24细胞,通过细胞增殖实验及克隆形成实验,检测莲心碱对T24细胞增殖的影响;应用Western Blot检测技术分析T24细胞所表达的p62蛋白、LC3B-Ⅱ蛋白的变化情况;采用激光共聚焦显微成像技术,观察莲心碱对T24细胞自噬体形成的影响。结果莲心碱能够明显抑制T24细胞的增殖,与空白组间的差异具有统计学意义(P<0.05,P<0.01),并呈量效依赖关系;Western Blot检测结果显示,莲心碱可增强p62蛋白与LC3B-Ⅱ蛋白的水平(P<0.05,P<0.01);激光共聚焦显微成像结果显示,莲心碱能够明显地诱导T24细胞自噬体形成(P<0.05,P<0.01)。结论莲心碱可明显抑制T24细胞的增殖,促进膀胱癌T24细胞的自噬体形成并增强p62与LC3B-Ⅱ蛋白的表达水平。Objective To investigate the effects of Liensinine on the proliferation of T24 cells and the formation of autophagosomes.Methods The T24 cells were treated with different concentrations of Liensinine,and the proliferation of T24 cells was determined by CCK-8 assay and clone formation assay.The expression levels of p62 and LC3 B-Ⅱ were analyzed by Western Blot;and the images of autophagosomes were collected by laser scanning confocal microscopy.Results The proliferation of T24 cells was significantly inhibited by Liensinine,and there was a significant difference(P <0.05,P <0.01) between the Liensinine groups and the control group in a dose-dependent manner.Western Blot assay results showed that Liensinine enhanced the expression of p62 and LC3 B-Ⅱ in T24 cells(P <0.05,P <0.01).The amount of autophagosomes in Liensinine group cells were more than control group which was observed by laser scanning confocal microscopy(P <0.05,P <0.01).Conclusion Liensinine significantly promoted the autophagosomes formation in T24 cells and enhanced the expression of p62 and LC3 B-Ⅱ.
