蓝刺头对绝经后骨质疏松症大鼠骨组织计量学及BMP-2、ADRβ-2R表达的影响

摘  要:目的观察蓝刺头对绝经后骨质疏松(PMOP)大鼠骨组织计量学及骨形态发生蛋白因子-2(BMP-2)和骨β2肾上腺素能受体(ADRβ-2R)表达的影响。方法将84只雌性Wistar大鼠随机分为模型组、假手术组、强骨胶囊组(69.2 mg·kg^(-1))、己烯雌酚组(0.046 mg·kg^(-1))及蓝刺头高、中、低剂量组(550、275、137.5 mg·kg^(-1)),每组12只。以摘除卵巢法建立PMOP大鼠模型。造模结束后,大鼠按上述剂量灌胃给药,每日1次,连续90 d。采用显微CT法检测骨组织计量学指标,即大鼠股骨干骺端骨密度(BMD)、骨小梁体积分数(BV/TV);采用免疫组织化学法检测股骨头BMP-2及ADRβ-2的表达。结果与假手术组比较,模型组大鼠的BMD、BV/TV以及BMP-2的表达水平均明显下降(P <0.05,P <0.01),而ADRβ-2R的表达水平显著升高(P <0.01)。与模型组比较,蓝刺头各剂量组的BMD、BV/TV水平明显提高(P <0.01),蓝刺头中剂量组的BMP-2表达水平明显上调(P <0.05),而ADRβ-2R的表达水平明显下调(P <0.05)。结论蓝刺头能改善PMOP大鼠的骨强度,增加骨量,促进骨形成、抑制骨吸收,其作用机制可能与上调BMP-2表达,下调ADRβ-2R表达有关。Objective To observe the effect of Echinops latifolius Tausch.on bone histomorphometry and expression of bone morphogenetic protein-2(BMP-2) and bone β2-adrenergic receptor(ADRβ-2 R) in rats with postmenopausal osteoporosis(PMOP).Methods Eighty-four Wistar rats were randomly divided into Sham operation group,model group,Echinops latifolius Tausch.high,medium and low dose groups(550,275,137.5 mg·kg-1),diethylstilbestrol group(0.046 mg·kg-1) and Qianggu capsule group(69.2 mg·kg-1).The model of PMOP was established by ovariectomy.After the model was established,the rats were given 20 mL·kg-1 of drugs by intragastric administration.The rats in Sham operation group and model group were given the same amount of normal saline once a day for 90 days.Bone density(BMD) and trabecular volume fraction of bone(BV/TV%) of the metaphysis of the right femur were measured by Micro-CT.The BMP-2 and ADRβ-2 R expressions of the left femoral head were detected by immunohistochemistry.Results Compared with Sham operation group,BMD,BV/TV% and BMP-2 were significantly decreased in model group(P <0.05,P <0.01),while ADRβ-2 R was significantly increased(P <0.01).Compared with model group,BMD and BV/TV% of Echinops latifolius Tausch.groups were significantly increased(P <0.01);BMP-2 expression in rats in the medium dose group was significantly increased(P <0.05),while ADRβ-2 R expression was significantly decreased(P <0.05).Conclusion Echinops latifolius Tausch.can improve bone strength,increase bone mass,promote bone formation and inhibit bone resorption.The mechanism mmay be related to increasing BMD,BV-TV%,up-regulation of BMP-2 and down-regulation of ADRβ-2 R expression.
