健脾理气、活血化瘀法促进慢性萎缩性胃炎大鼠胃腺体增殖的作用机制

摘　　要：目的:探讨健脾理气、活血化瘀法促进慢性萎缩性胃炎(CAG)大鼠胃腺体增殖的作用机制。方法:将健康Wistar大鼠随机分为空白对照组、造模组。造模组大鼠采用复合造模法制备CAG大鼠模型。造模成功后将模型大鼠随机分为模型组、中药低剂量组、中药高剂量组、胃复春组,分别给药30 d。免疫组化方法测定胃组织IL-11、PCNA、TGF-β1、RegI。结果:中药高、低剂量组大鼠胃组织IL-11、TGF-β1表达均低于模型组及胃复春组;中药高剂量组大鼠胃组织PCNA、RegI表达均高于模型组及胃复春组;中药低剂量组大鼠胃组织PCNA的表达高于模型组,低于胃复春组;中药低剂量胃组织大鼠RegI表达高于模型组,与胃复春组相仿。结论:健脾理气、活血化瘀法对CAG大鼠模型具有治疗作用,可降低CAG大鼠模型胃组织IL-11、TGF-β1表达,提高PCNA、RegI表达。Objective:To explore the mechanism of promoting the proliferation of gastric glands in rats with chronic atrophic gastritis(CAG)by fortify the spleen and regulate qi,activate blood and resolve stasis method.Methods:Healthy Wistar rats were randomly divided into a blank control group and a model group.CAG rat model was prepared by compound modeling method.After successful modeling,the model rats were randomly divided into a model group,Chinese medicine low-dose group,Chinese medicine high-dose group,and Weifuchun group,which were administered for 30 days.The levels of IL-11,PCNA,TGF-β1 and RegI in gastric tissue were measured by immunohistochemistry.Results:The expressions of IL-11 and TGF-β1 in gastric tissue of rats in Chinese medicine high and low-dose groups were lower than those in model group and Weifuchun group.The expressions of PCNA and RegI in gastric tissue of rats in Chinese medicine high-dose group were higher than those in model group and Weifuchun group.The expression of PCNA in gastric tissue of rats in Chinese medicine low-dose group was higher than that in model group and lower than that in Weifuchun group.The expression of RegI in gastric tissue of rats in Chinese medicine low-dose group was higher than that in model group,similar to that in Weifuchun group.Conclusion:The method of fortify the spleen and regulate qi,activate blood and resolve stasis has therapeutic effect on CAG rat model,which can reduce the expression of IL-11 and TGF-β1 in gastric tissue,and increase the expression of PCNA and RegI.
