摘 要:目的:探讨滋肾平肝方对二肾一夹高血压大鼠(2K1C-RHR)肾损害的保护作用及对TGFβ1/Smads信号通路的影响。方法:复制2K1C-RHR模型,造模后4周评估造模情况,选取造模成功的32只大鼠随机分为模型组,滋肾平肝方高、低剂量组,厄贝沙坦组,另选8只同龄SD大鼠为假手术组,各组大鼠连续灌胃给药8周,分别于造模4周,灌胃第2、4、6、8周检测大鼠尾动脉收缩压;给药结束后检测大鼠尿微量白蛋白(mALb)、β2-微球蛋白(β2-MG)、N-乙酰-β氨基葡萄糖苷酶(NAG)、血肌酐(SCr)、尿素氮(BUN)、胱抑素C(CysC)含量;观察肾组织病理形态学改变;蛋白质印迹法(Western blotting法)检测TGFβ1、p-Smad2/3蛋白表达。结果:滋肾平肝方高剂量组、厄贝沙坦组血压均明显低于模型组(P<0.05,P<0.01);滋肾平肝方高剂量组尿mALb、β2-MG、NAG,血清SCr、CysC均明显降低,与模型组比较,差异有统计学意义(P<0.05,P<0.01);滋肾平肝方高剂量组肾组织病理损伤程度明显减轻;滋肾平肝方高剂量组、厄贝沙坦组肾组织TGFβ1、p-smad2/3蛋白表达均明显降低,与模型组比较,差异有统计学意义(P<0.01)。结论:滋肾平肝方对高血压肾损害具有保护作用,其机制可能与抑制TGFβ1/Smads信号通路有关。Objective:To explore the protective effect of Zishen Pinggan Recipe on renal damage in twokidney-one-clip hypertensive rats(2 K1 C-RHR)and its influence on TGFβ1/Smads signaling pathway.Methods:The 2 K1 C-RHR model was replicated,and evaluate the modeling situation 4 weeks after making the model.A total of 32 rats successfully modeled were randomly divided into model group,Zishen Pinggan Recipe low-dose group,Zishen Pinggan Recipe high-low dose group,irbesartan group,and 8 SD rats of the same age as sham operation group.The rats in each group were given intragastric administration continuously for 8 weeks,and the rats’tail artery systolic blood pressure was measured at the 2,4,6,and 8 week after the model was established.At the end of administration,the contents of urinary microalbumin(mAlb),β2-microglobulin(β2-mg),N-acetyl-β-Glucosaminidase(NAG),serum creatinine(SCr),urea nitrogen(BUN)and cystatin C(CysC)were detected.Observe the pathological changes of kidney tissue.The expression of TGFβ1 and p-Smad2/3 was detected by Western blotting.Results:The blood pressure of Zishen Pinggan Recipe high-dose group and irbesartan group were significantly lower than that of model group(P<0.05 or P<0.01).Compared with the model group,the urine mAlb,β2-mg,NAG,SCr and CysC in the Zishen Pinggan Recipe high-dose group were significantly decreased(P<0.05 or P<0.01).The kidney tissue pathological damage was significantly reduced in the Zishen Pinggan Recipe high-dose group.Compared with the model group,the expression of TGFβ1 and p-Smad2/3 protein in kidney tissue of Zishen Pinggan Recipe high-dose group and irbesartan group were significantly lower than those in model group(P<0.01).Conclusion:Zishen Pinggan Recipe has a protective effect on hypertensive renal damage,and its mechanism may be related to the inhibition of TGFβ1/Smads signaling pathway.
