白头翁汤中小檗碱在正常与溃疡性结肠炎模型大鼠组织分布比较研究

摘　　要：目的:探讨小檗碱在溃疡性结肠炎模型大鼠与正常大鼠体内组织分布的差异。方法:将雄性SD大鼠随机分为正常组和溃疡性结肠炎模型组,灌胃给予白头翁汤水煎液,于1、3、5、7、12 h麻醉正常和模型大鼠,取小肠、肝、肺、脾、肾、心等组织,各组织样品分别经甲醇沉淀蛋白后,采用UPLC-ESI-MS/MS分析方法测定各组织中小檗碱的浓度。结果:小檗碱在大鼠体内存在广泛分布与代谢。与正常组大鼠比较,模型组大鼠小檗碱的含量在小肠、肝和肺于1 h明显升高(P<0.05);在小肠、肝、肺、脾、肾和心于3 h明显升高(P<0.05);在小肠、肝、肺和肾于5 h明显升高(P<0.05);在小肠、肝和脾于7 h明显升高(P<0.05);在小肠、心于12 h明显升高(P<0.05)。结论:在溃疡性结肠炎状态下,大鼠灌胃白头翁汤后小檗碱的组织分布存在显著差异。Objective:To investigate the difference in tissue distribution of berberine in ulcerative colitis model rats and normal rats.Methods:Male SD rats were randomly divided into normal group and ulcerative colitis model group,and the Baitouweng Decoction was given by gavage.Normal and model rats were anesthetized at 1,3,5,7,and 12 h,and the small intestine and liver,lung,spleen,kidney,heart and other tissues were collected.After each tissue sample was precipitated with methanol,the UPLC-ESI-MS/MS analysis method was used to determine the concentration of berberine in each tissue.Results:Berberine is widely distributed and metabolized in rats.Compared with the normal group,the content of berberine in small intestine,liver and lung was significantly increased at 1 h(P<0.05),in small intestine,liver,lung,spleen,kidney and heart at 3 h(P<0.05),in small intestine,liver,lung and kidney at 5 h(P<0.05),in small intestine,liver and spleen at 7 h(P<0.05),in small intestine and heart at 12 h(P<0.05).Conclusion:There is a significant difference in the tissue distribution of berberine after oral administration of Baitouweng Decoction in rats with ulcerative colitis.
