摘 要:目的探讨研究脐带间充质干细胞(ucMSCs)对大鼠创伤性胰腺炎(TP)的治疗保护作用。方法 30只健康成年雄性SD大鼠,随机分为Sham组、TP组、ucMSCs治疗组,每组10只。采用自主研发的基于损伤范围控制的胰腺创伤动物模型建立TP大鼠模型,造模结束后72 h取材,行HE染色评估胰腺组织损伤程度,ELISA法检测血清中淀粉酶、脂肪酶和炎症因子的含量。结果造模后,胰腺组织病理损伤明显,血清淀粉酶、血清脂肪酶和炎症因子表达含量均明显升高;相比TP组,ucMSCs治疗组胰腺组织病理学评分明显降低(P <0.05),血清淀粉酶、血清脂肪酶浓度有下降趋势(P <0.05),血清炎症因子IL-6和TNF-α浓度也相对降低(P <0.05)。结论 ucMSCs移植能有效减轻大鼠创伤性胰腺炎胰腺病理损伤及全身炎症反应,对大鼠创伤性胰腺炎具有一定的保护作用。Objective To explore the therapeutic and protective effects of umbilical cord mesenchymal stem cells(ucMSCs) on traumatic pancreatitis in rats. Methods A total of 30 healthy adult male SD rats were selected and randomly divided into the Sham group, TP group, and ucMSCs treatment group, with 10 each. A rat model of TP was established using the self-developed animal model of pancreatic trauma based on control of the trauma area. Sampling was conducted 72 hours after the modeling, then HE staining was performed to evaluate the extent of injury of pancreatic tissues, and the levels of serum amylase, serum lipase, and inflammatory cytokines were detected using the ELISA method. Results After modeling, the pathological injury of pancreatic tissues was obvious, and the expression levels of serum amylase, serum lipase, and inflammatory cytokines were significantly increased.Compared with the TP group, the pathological scores of pancreatic tissues in the ucMSCs treatment group were significantly decreased(P < 0.05), the concentrations of serum amylase and serum lipase showed a downward trend(P < 0.05), and the concentrations of serum inflammatory cytokines IL-6 and TNF-α were also relatively decreased(P < 0.05). Conclusion The transplantation of ucMSCs can effectively reduce the pathological injury of pancreatic tissues and systemic inflammatory response for traumatic pancreatitis in rats, and has a certain protective effect on traumatic pancreatitis in rats.
