摘 要:近年来H9N2亚型禽流感病毒(AIV) h9.4.2.5分支在我国广泛流行。为了解广东地区H9N2 AIV的遗传与变异特点及进化规律,采用RT-PCR技术对自2018-2019年分离到的5株H9N2 AIV的HA与NA基因进行扩增、测序和序列比对。结果显示,5株病毒分离株的HA基因在遗传进化树中依然均属于h9.4.2.5分支,NA基因均属于Y280-like分支;分离到的5株H9N2 AIV在HA裂解位点处均只有1个碱性氨基酸(R),与低致病性AIV的特征相符;所有菌株受体结合位点第234位氨基酸是亮氨酸(L),显示出典型的人流感病毒受体结合特性;分离株均缺乏218~220位糖基化位点,同时增加了313~315位糖基化位点。研究表明流行毒株在不同程度上发生了基因重组,导致与现有疫苗的抗原匹配存在差异,因此要积极关注禽流感防控中出现的新问题。In recent years,the H9 N2 avian influenza virus has been spread quickly to most regions of the China.The sequencing and phylogenetic analysis of the hemagglutinin gene of the isolation strains showed that most isolation strains were clustered to h9.4.2.5 lineage.In order to explore the genetic and variability and evolution of H9 N2 avian influenza virus(AIV) in Guangdong,we used RT-PCR to clone the HA and NA genes of five H9 N2 AIV isolated from 2018 to 2019.The results showed that the HA genes of the five strains of virus still belong to the h9.4.2.5 lineage,and the NA genes belong to the Y280-like lineage.No consecutive basic amino acids insertion was found in the HA cleavage sites,showing the feature of low-pathogenic AIV.The 234 receptor binding sites of all strains were L(leucine),showing typical human influenza virus receptor binding properties.In addition,the isolates lacked the 218 to 220 glycosylation sites and added 313 to 315 glycosylation sites.These results indicated that the dynamic evolution of H9 N2 in the field emphasizes the need for monitoring of the situation in terms of H9 N2 AIV in Guangdong.So it is necessary to actively pay attention to new challenger in the prevention and control of avian influenza.
