高能低蛋白日粮诱导的脂肪肝出血综合征(FLHS)蛋鸡脂肪组织代谢差异的比较

摘　　要：为了比较高能低蛋白日粮诱导的脂肪肝出血综合征(FLHS)蛋鸡脂肪组织代谢的差异,将海兰褐蛋鸡分为对照组与病理组,每组40只,对照组饲喂基础日粮,病理组饲喂高能低蛋白日粮。饲养80 d后,采集肝脏和脂肪组织进行HE染色观察,同时测定血清中白细胞介素6(IL-6)的水平并通过实时荧光定量PCR测定IL-6在脂肪组织中的表达差异变化。HE染色结果表明,通过给予高能低蛋白日粮可成功建立FLHS模型;所建立FLHS组(病理组)脂肪组织出现脂质代谢异常,其脂滴(LDs)直径明显(P<0.01)大于对照组LDs。FLHS组中IL-6在脂肪组织中的表达水平与血清水平均显著高于(P<0.05或P<0.01)对照组,提示患FLHS的蛋鸡脂肪组织代谢失常导致机体IL-6水平升高,从而对肝脏的肿大、出血等症状呈正向诱导。In order to compare the difference in fat tissue metabolism of fatty liver hemorrhage syndrome(FLHS) layer hens induced by high-energy low-protein diets,Hailan brown layer hens were divided into a control group and a model group,with 40 animals in each group.The control group was fed a basic diet.The pathological group was fed a high-energy low-protein diet.After 80 days of feeding,the liver and adipose tissue were collected for HE staining;at the same time,the level of interleukin 6(IL-6) in serum and its expression in adipose tissue was measured by real-time fluorescence quantitative PCR amplification.HE staining results showed that the FLHS model was successfully established by giving high energy and low protein diets;the established FLHS group had abnormal lipid metabolism in the adipose tissue,and the size of lipid droplets(LDs) was significantly(P<0.01) larger than that of the control group.The expression level of IL-6 in adipose tissue and serum level in FLHS group were significantly higher(P<0.05 or P<0.01) than the control group,suggesting that abnormal metabolism of adipose tissue in laying hens with FLHS lead to increase IL-6 levels in the body,thereby positively inducing symptoms such as liver enlargement and bleeding.
