HPLC法测定蒙药嘎扎古瓦中木香烃内酯和去氢木香内酯的含量

摘　　要：建立嘎扎古瓦中木香烃内酯和去氢木香内酯的含量测定方法。高效液相色谱法,Pheomenex-C18(250mm×4.6mm,5μm)色谱柱,流动相为甲醇-水(65∶35);流速1.0 mL·min^-1;柱温30℃;进样量为10μL;检测波长为225nm。木香烃内酯在0.206~2.060μg范围内呈良好的线性关系(r=0.9999),去氢木香内酯在0.166~1.658μg范围内呈良好的线性关系(r=0.9999);木香烃内酯平均加样回收率为96.57%(RSD=1.95%),去氢木香内酯平均加样回收率为95.69%(RSD=1.96%)。该法简便、准确、专属性好,可用于嘎扎古瓦中木香烃内酯和去氢木香内酯的测定。To establish an HPLC method for determination of costunolide and dehydrocostus lactone in Gazhaguwa.The Pheomenex-C18 column(250mm×4.6 mm,5μm)was adopted.The mobile phase consisted of methanol-water(65∶35);the flow rate was 1.0 mL·min^-1;the column temperature was 30℃;the injection amount was 10μL;the detection wavelength was 225 nm.The costunolide had a good linear relationship with the injection amount ranged from 0.206 to 2.060μg(r=0.9999).The dehydrocostus lactone had a good linear relationship with the injection amount ranged from 0.166 to 1.658μg(r=0.9999).The average recovery of costunolide was 96.57%(RSD=1.95%).The average recovery of dehydrocostus lactone was 95.69%(RSD=1.96%).This method is simple,accurate and specific,which can be used for the determination of costunolide and dehydrocostus lactone in Gazhaguwa.
