彩叶矾根‘草莓漩涡’组培再生体系建立及快繁技术研究

摘　　要：以矾根品种‘草莓漩涡’一年生苗的茎尖、叶片和叶柄为外植体,以MS为基本培养基,添加不同浓度BA、KT、NAA、IBA、IAA,研究不同种类和浓度激素对外植体诱导丛生芽、继代增殖和试管苗生根的影响,并进行试管苗炼苗和移栽基质试验。结果表明:3个外植体部位均能诱导出丛生芽,诱导丛生芽数量最多的是叶片,最佳丛生芽诱导培养基为MS+6-BA 0.5 mg/L+NAA 0.1 mg/L,平均诱导率达到81.2%,增殖系数平均可达9;最佳生根培养基配方为1/2 MS+IBA 0.1 mg/L,生根率达100%;最佳组培苗炼苗基质为珍珠岩,移栽基质为草炭+珍珠岩(3∶1)。矾根的叶片、叶柄是外植体的最好选择,材料丰富,表面杀菌容易,在短期内能获得大量组培苗。Take the shoot tips,leaves and Petioles of the one-year-old Heuchera micrantha’strawberry Swirl’as explants,and MS as the basic medium with different concentrations of BA,KT,NAA,IBA,IAA,the effects of different kinds and concentrations of hormones on adventitious bud induction,subculture multiplication and rooting of test-tube plantlets were studied.The results showed that cluster buds could be induced from all three explants,the number of cluster buds was the most,the best medium was MS+6-BA 0.5 mg/L+NAA 0.1 mg/L,the average induction rate was 81.2%,the best rooting medium was 1/2 ms+IBA 0.1 mg/L,the rooting rate was 100%,the best medium for tissue culture seedling was perlite,the best medium for transplanting was peat+Perlite(3:1).Leaves and petioles are the best choice of explants,material-rich,easy surface sterilization,can obtain a large number of tissue culture seedlings in a short period of time.
