摘 要:目的研究盐酸小檗碱对溃疡性结肠炎大鼠炎症及结肠组织紧密连接蛋白occludin表达的影响。方法将60只SPF级Wistar大鼠随机分为空白组、模型组、实验组及对照组,各15只。模型组、实验组及对照组大鼠均采用连续饮用3%葡聚糖硫酸钠溶液7 d法构建溃疡性结肠炎大鼠模型,空白组饮用蒸馏水。建模成功后实验组灌胃150 mg·kg-1·d-1盐酸小檗碱,对照组灌胃柳氮磺吡啶片30 mg·kg-1·d-1,空白组与模型组则灌胃等量生理盐水,连续干预14 d。干预后对大鼠进行疾病活动系数(DAI)评估;以苏木精-伊红(HE)染色观察大鼠结肠组织病理学形态,进行结肠组织大体形态损伤指数(CMDI)评分及结肠组织病理学评分;以酶联免疫吸附(ELISA)法检测结肠组织炎性细胞因子含量;以免疫组化法检测结肠组织紧密连接蛋白occludin表达情况。结果空白组、模型组、实验组及对照组大鼠结肠组织肿瘤坏死因子-α(TNF-α)含量分别为(64.31±12.56),(279.14±19.58),(184.01±21.33),(179.85±19.67)pg·mL-1;白细胞介素-1β(IL-1β)含量分别为(72.84±9.21),(238.17±13.04),(143.55±13.01),(140.69±12.58)pg·mL-1;结肠组织紧密连接蛋白occludin蛋白阳性细胞百分比分别为(66.87±12.46)%,(12.57±3.21)%,(55.46±11.23)%,(62.43±10.79)%。实验组及对照组大鼠DAI评分、CMDI评分、病理学评分和结肠组织TNF-α、IL-1β含量均较模型组降低,结肠组织紧密连接蛋白occludin蛋白阳性细胞百分比较模型组升高,且实验组与模型组上述指标差异均有统计学意义(均P<0.05),且对照组结肠组织紧密连接蛋白occludin蛋白阳性细胞百分比较实验组升高(P<0.05),而其他指标2组间差异均无统计学意义(均P>0.05)。结论盐酸小檗碱可有效抑制结肠炎大鼠局部炎性反应,缓解结肠组织炎性病理损伤,其作用机制可能与其显著上调结肠组织紧密连接蛋白occludin表达相关。Objective To explore the effect of berberine hydrochloride on the inflammation and colon tissue tight junction protein occludin expression in ulcerative colitis rats.Methods Sixty SPF Wistar rats were divided randomly into blank group,model group,test group and control group,15 rats in each group.Rats in model group,test group and control group were built ulcerative colitis rat model by drinking 3%dextran sodium sulfate solution for 7 d,blank group treated with drinking distilled water.After successful modeling,test group was treated with 150 mg·kg-1·d-1 berberine hydrochloride by gavage,control group was treated with 30 mg·kg-1·d-1 sulfasalazine tablets by gavage blank group and model group were treated with the same amount of normal saline by gavage.All groups were intervented for 14 d.After intervened,the disease activity index(DAI)of rats were evaluated;the colon tissue pathology morphology was observed by hematoxylin and eosin(HE)staining,the general shape colon tissue damage index(CMDI)score and colon histopathological grading were evaluated;the colon tissue inflammatory cytokine levels were detected by enzyme-linked immunosorbent assay(ELISA);the colon tissue tight junction protein occludin expression was detected by immunohistochemical method.Results The tumor necrosis factor-α(TNF-α)in colon tissue of rats in blank group,model group,test group and control group were(64.31±12.56),(279.14±19.58),(184.01±21.33),(179.85±19.67)pg·mL-1;interleukin-1β(IL-1β)were(72.84±9.21),(238.17±13.04),(143.55±13.01),(140.69±12.58)pg·mL-1;the colon tissue tight junction protein occludin positive cell proportions were(66.87±12.46)%,(12.57±3.21)%,(55.46±11.23)%,(62.43±10.79)%.The DAI score,CMDI score and colon histopathological grading,colon tissue TNF-α,IL-1βcontents in test group and control group were lower than those in model group,the colon tissue tight junction protein occludin positive cell proportion were higher than those in model group,and there was significant difference of the above indexes
