摘 要:根据伪三元相图及星点设计-效应面法筛选出蒲公英根提取物自微乳给药系统的最佳处方。根据蒲公英根提取物在油相、乳化剂及助乳化剂中的溶解度,分别筛选出较合适的油相、乳化剂及助乳化剂,运用伪三元相图法初步筛选空白自微乳处方,再以不同蒲公英根提取物自微乳处方中的咖啡酸载药量、总黄酮载药量和粒径为指标,采用星点设计-效应面优化法,确定蒲公英根提取物自微乳最佳处方。显示蒲公英根提取物在油相(薄荷油)、乳化剂(吐温80)与助乳化剂(1,2-丙二醇)中溶解度较大。乳化剂吐温80与助乳化剂1,2-丙二醇Km值为1时,空白自微乳所形成的微乳在伪三元相图中有最大区域面积,当油相∶乳化剂∶助乳化剂=1∶2∶2时空白自微乳形成微乳区域面积最大。星点设计-效应面法得出蒲公英根提取物自微乳优化处方为油相比例为35.61%,Km值为1.58,咖啡酸载药量21.98 mg/g,总黄酮载药量26.34 mg/g,平均粒径51 nm。表明该处方设计方法简单,星点设计-效应面法预测性良好,结果可靠,性质稳定,为进一步开发与研究奠定基础。Based on the pseudo ternary phase diagram and central composite design response surface methodology,the best formulation of self microemulsion drug delivery system of dandelion root extract was screened.According to the solubility of dandelion root extract in oil phase,emulsifier and co-emulsifier,the suitable oil phase,emulsifier and co-emulsifier were selected,respectively.The blank self microemulsion formulation was preliminarily screened by pseudo ternary phase diagram method.Then,the caffeic acid loading,total flavonoids loading and particle size of different self microemulsion formulations of dandelion root extract were taken as indexes,and the central composite design response surface optimization was adopted.The best formulation of self microemulsion of dandelion root extract was determined by chemical method.The results showed that the solubility of dandelion root extract was higher in the oil phase,i.e.peppermint oil,Tween 80,and co-emulsifier,l,2-propanediol.When the Km value of Tween 80 and co-emulsifier 1,2-propanediol was 1,the area of microemulsion formed by blank self microemulsion was the largest in the pseudo ternary phase diagram,and the area of microemulsion formed by white self microemulsion was the largest when the oil phase∶emulsifier∶co emulsifier=1∶2∶2.The optimized formulation of self microemulsion of dandelion root extract by central composite design response surface methodology was as follows:oil phase ratio was 35.61%,Km value was 1.58,caffeic acid loading was 21.98 mg/g,total flavonoids loading was 26.34 mg/g,and the average particle size was 51 nm.The results show that the prescription design method is simple,and the central composite design response surface method has good predictability,reliable results and stable properties,which will lay a foundation for further development and study.
