保幼激素诱导尖唇散白蚁兵蚁分化的特征及其相关基因的表达谱

摘  要:为明确保幼激素调控白蚁兵蚁分化的机理,以尖唇散白蚁Reticulitermes aculabialis为研究对象,用保幼激素Ⅲ及其类似物吡丙醚诱导其工蚁向兵蚁分化,并用荧光定量PCR技术对兵蚁分化过程中保幼激素信号相关基因的表达模式进行分析。结果显示,春、夏季节保幼激素Ⅲ和吡丙醚对尖唇散白蚁前兵蚁的诱导率和死亡率均高于秋、冬季节,如在春、夏季的诱导率最高达60.84%,死亡率最高为47.18%;而在秋、冬季的诱导率最高仅为20.00%,死亡率最高为12.50%。相同浓度的保幼激素Ⅲ和吡丙醚诱导尖唇散白蚁时,在前兵蚁阶段Met、Kr-h1、Br-C和USP基因的表达模式基本相似,除了USP外,均有表达高峰;Met在前兵蚁阶段的第5天达到高峰;保幼激素Ⅲ诱导与吡丙醚诱导相比,Kr-h1和Br-C的表达高峰均提前2 d。不同浓度的吡丙醚诱导白蚁时,120μg/mL和160μg/mL浓度诱导下Met、Kr-h1和Br-C基因均在第5天出现表达高峰(Br-C的160μg/mL处理除外,表达高峰推迟2 d)。表明春、夏季保幼激素及其类似物更容易诱导分化出前兵蚁,且保幼激素Ⅲ比吡丙醚对下游基因的影响更明显。In order to explore the mechanism of termite soldier differentiation regulated by juvenile hormone, Reticulitermes aculabialis was used to investigate the differentiation of worker-to-soldier by using juvenile hormone Ⅲ and its analogue-pyripropoxyfen, and expression profiling of juvenile hormone signal-related genes was analyzed using real-time fluorescence quantitative PCR during the pre-soldier phase. The results showed that the inductivity and mortality of pre-soldier by juvenile hormone Ⅲ and pyripropoxyfen in spring and summer were higher than that in autumn and winter: the highest inductivity and mortality were 60.84% and 47.18%, respectively, in spring and summer, while those were only20.00% and 12.50%, respectively, in autumn and winter. The expression patterns of the genes Met, Krh1, Br-C and USP were similar, except for USP, when the pre-soldiers were induced with the same concentration of juvenile hormone Ⅲ and pyripropoxyfen. The peak of Met was observed on the 5 th day of pre-soldier;in contrast, the expression peaks of Kr-h1 and Br-C appeared two days earlier. With pyripropoxyfen inducing workers at different concentrations, the expression peaks of Met, Kr-h1 and Br-C were observed at the 5 th day of pre-soldier in 120 μg/mL and 160 μg/mL groups(except for Br-C in 160μg/mL group, which was delayed for two days). These results indicated that juvenile hormone and its analogue induced pre-worker more easily in spring/summer than in autumn/winter, and juvenile hormone Ⅲ imposed a more obvious impact on downstream genes than that of pyripropoxyfen.
