摘  要:目的研究在人表皮细胞中hsa-let-7b-5p的作用和其对ΔNp63表达变化的影响,探讨hsa-let-7b-5p如何影响人表皮细胞的生物功能。方法人角质形成细胞株(HaCaT)分别转染hsa-let-7b-5p mimic NC、hsa-let-7b-5p mimic、hsa-let-7b-5p inhibitor和hsa-let-7b-5p inhibitor NC,通过CCK8检测细胞增殖,Transwell实验检测细胞迁移和细胞侵袭,利用miRcode网站和RNAhybrid网站预测p63和hsa-let-7b-5p结合,利用实时荧光定量PCR检测hsa-let-7b-5p及p63表达情况。结果hsa-let-7b-5p的高表达对HaCaT细胞的增殖、迁移和侵袭均有抑制作用,hsa-let-7b-5p表达的抑制则促进了HaCaT细胞的增殖、迁移和侵袭。通过网站预测发现hsa-let-7b-5p可与对皮肤表皮细胞有重要作用的p63结合;实时定量PCR分析发现hsa-let-7b-5p影响p63的表达。结论hsa-let-7b-5p可能通过靶向p63 mRNA水平而在人表皮细胞中发挥作用。Objective To study the role of microRNA hsa-let-7b-5p in human epidermal cells and its effect onΔNp63 expres-sion,in order to investigate how hsa-let-7b-5p regulates biological functions of the human epidermal cells.Methods The human kerat-inocyte line(HaCaT)was transfected with hsa-let-7b-5p mimic NC,hsa-let-7b-5p mimic,hsa-let-7b-5p inhibitor and hsa-let-7b-5p in-hibitor NC.The cell proliferation was detected by cck8 assay,cell migration and cell invasion were detected by Transwell experiments.MiRcode and RNAhybrid websites were used to predict the binding of p63 and hsa-let-7b-5p.The real-time fluorescence quantitative PCR was used to detect the expression of hsa-let-7b-5p and p63.Results The relative high expression of hsa-let-7b-5p inhibited the proliferation,migration and invasion of HaCaT cells,while the inhibition of hsa-let-7b-5p expression promoted the proliferation,mi-gration and invasion of HaCaT cells.The website predicted that hsa-let-7b-5p can bind to p63,which plays an important role in skin epidermal cells.The results of real-time quantitative PCR revealed that hsa-let-7b-5p affected the expression of p63.Conclusion hsa-let-7b-5p possibly plays a role in HaCaT cells by targeting the mRNA level of p63.
