摘  要:氧气对生命至关重要,低氧诱导因子(hypoxia inducible factor,HIF)通路的发现揭示了机体适应氧气变化的新机制。缺氧情况下,机体感受到氧气不足,HIF像一个开关,激活体内众多基因转录,使机体适应低氧环境。由于许多疾病与缺氧相关,对HIF的认识也可能为许多疾病的治疗提供了全新的思路和机遇。目前针对HIF通路研发的HIF稳定剂已应用于临床肾性贫血的治疗。考虑到HIF调节的下游靶基因在包括能量代谢、细胞保护、脂质代谢、血压调节和血管生成等病理生理过程均具有重要作用,靶向调节HIF通路还可能为炎性贫血、缺氧性肾脏病、慢性肾脏病相关心血管疾病以及其他缺氧性疾病的治疗提供新的启发。文章对HIF通路多重作用及其临床意义进行概述。Oxygen is essential to life.The discovery of hypoxia inducible factor(HIF)pathway reveals the new mechanism of body adapting to the the changes in oxygen.The body senses the deficiency in oxygen under hypoxia,and HIF may act as a switch,transcriptionally activating many genes in the body exposed to hypoxia,making the body adapting to the hypoxic environment.Therefore,this innovative discovery provides a novel theoretical basis for the research in many related fields.Currently,HIF stabilizers have been used in the clinical treatment of renal anemia.Giving that the ability of HIF activation to induce endogenous defense mechanisms against hypoxia,including metabolic reprogramming,cytoprotection,lipid metabolism,blood pressure regulation and angiogenesis,has provided a strong rationale for effective treatment of anemia with inflammation,ischemic kidney disease,chronic kidney disease-related cardiovascular disease and other hypoxic diseases.Here,we discussed the multiple effects of HIF pathway and their potential clinical implications.
