摘  要:为探讨MDR1基因沉默对紫杉醇诱导HT9急性早幼粒白血病耐药细胞凋亡的影响。本研究将已构建的干涉MDR1基因表达载体转染HT9细胞,通过流式细胞术、MTT法分析HT9细胞对药物的外排能力和对紫杉醇的敏感性,电镜、荧光显微镜、流式细胞术检测紫杉醇诱导细胞凋亡的形态学、细胞周期进程变化。结果显示,干涉MDR1基因后,HT9细胞内Rh123荧光强度增加,细胞外排能力减弱,细胞对紫杉醇的敏感性增强,促进紫杉醇诱导耐药细胞呈现细胞凋亡表型,促使细胞周期阻滞在S和G2/M期。说明MDR1基因沉默能增强紫杉醇诱导HT9细胞凋亡,为逆转白血病耐药、提高白血病化疗效果提供参考依据。To investigate the effects of MDR1 gene silencing on apoptosis of HT9 cells induced by paclitaxel.In this study,the constructed MDR1 gene expression vector was transfected into HT9 cells.The efflux function and sensitivity of cells to paclitaxel were analyzed by flow cytometry and MTT assay.The effects of paclitaxel on the cell morphology and cell cycle process were detected through the electron microscope,fluorescent microscope and flow cytometry.The results showed that the fluorescence intensity of Rhodamine 123 in cells was increased,the efflux function of cells was reduced,and the drug sensitivity of cells to paclitaxel was enhanced after MDR1 gene was silenced.MDR1 gene silencing promoted paclitaxel-induced apoptotic phenotype in drug-resistant cells,and arresting the cell cycle in S and G2/M stage.It is indicated that MDR1 gene silencing enhanced the sensitivity of HT9 cells to paclitaxel.This study provides a reference for reversing the drug resistance of leukemia and improving the effect of chemotherapy in leukemia.
