摘 要:为了明确胡杨Pe BZR1基因对盐胁迫的响应,本研究克隆胡杨PeBZR1基因的cDNA序列,利用农杆菌介导法,将过表达载体pBI121-35S::PeBZR1转化野生型烟草,并通过筛选获得过表达T2代的转基因植株。对转基因植株和野生型植株进行以下处理:对照(Control)、100 mmol/L NaCl处理以及200 mmol/L NaCl处理。结果如下:在正常生长条件下,转基因烟草植株的长势更好,其根长和叶总面积较野生型分别增加了28.74%、17.06%;在100 mmol/L Na Cl的处理条件下,转基因烟草的根长和叶总面积较野生型相比没有显著性的差异,但转基因叶片细胞损伤程度、叶片内的H2O2和O2-的积累水平均低于野生型;在200 mmol/L NaCl条件下,转基因烟草根长和叶总面积较野生型相比分别平均增加了15.41%、65.30%,且转基因叶片细胞损伤程度、叶片内的H2O2和O2-的积累水平均显著低于野生型,并表现出较强的耐盐能力。以上结果表明,异源过表达PeBZR1基因可以促进转基因植株在正常条件下的生长,并提高转基因植株对高盐的耐受性。In order to clarify the response of Populus euphrates PeBZR1 gene to salt stress,we cloned the cDNA sequence of Populus euphrates Pe BZR1 gene,transformed the overexpressed vector pBI121-35 s::PeBZR1 into wild-type tobacco by Agrobacterium-mediated method,and transgenic plants with overexpression of T2 generation were obtained by screening.Transgenic plants and wild-type plants were treated as follows:Control,100 mmol/L NaCl treatment and 200 mmol/L NaCl treatment.The results were as follows:under normal growth conditions,the growth of transgenic tobacco plants was better,and the total root length and leaf area increased by 28.74%and17.06%respectively compared with the wild type.Under the condition of 100 mmol/L NaCl treatment,there was no significant difference in the root length and total leaf area of transgenic tobacco compared with that of the wild type.However,the damage degree of transgenic leaf cells and the accumulation level of H2O2 and O2-in the leaves were all lower than that of the wild type.Under the condition of 200 mmol/L NaCl,the root length and total leaf area of transgenic tobacco increased by 15.41%and 65.30%respectively on average compared with that of the wild type,and the degree of leaf cell damage and the accumulation level of H2O2 and O2-in the leaves were significantly lower than that of the wild type,and showed strong salt tolerance.These results indicate that heterogenous overexpression of PeBZR1 gene can promote the growth of transgenic plants under normal conditions and enhance the tolerance of transgenic plants to high salt stress.
