摘  要:探究Bcl-2基因在不同妊娠时期胎盘滋养细胞中的表达,明确Bcl-2基因与胎盘滋养细胞凋亡的相关性。采集在皖北煤电集团总医院妇产科住院分娩的早、中、晚期妊娠妇女的胎盘各15例。用TUNEL法检测滋养层细胞的凋亡,免疫组织化学法检测Bcl-2和Bax蛋白在滋养层细胞的表达位置及表达量,免疫印迹法(Western blotting)和实时荧光定量PCR(qPCR)法分别检测Bcl-2和Bax蛋白及mRNA的表达。结果显示,凋亡细胞核主要见于合体滋养细胞中,而细胞滋养细胞中很少检测到凋亡,细胞凋亡发生的频率与胎龄呈负相关。Bcl-2和Bax蛋白在各妊娠期合体滋养细胞中均有表达,但在细胞滋养细胞及其他部位未见表达。Bcl-2和Bax蛋白表达水平与胎龄呈正相关,晚期胎盘中Bcl-2和Bax蛋白的表达明显高于早期胎盘(p<0.05)和中期胎盘(p<0.05)。Bcl-2 mRNA和Bax mRNA表达水平与胎龄呈正相关,早期、中期、晚期胎龄间的mRNA表达水平存在显著差异(p<0.05)。Bcl-2和Bax对滋养层细胞,尤其是合体滋养细胞的凋亡可能起到重要的调节作用,其机制可能是在晚期胎盘中发挥抵抗细胞凋亡的作用,以此维持晚期胎盘中滋养层细胞的数量,保证正常的胎盘功能。In this study,we want to investigate the relationship between Bcl-2 gene and human placental trophoblast apoptosis by exploring the expression of Bcl-2 gene in placental trophoblast cells at different stages of pregnancy.45 cases of placenta from first,middle and term pregnant women(15 each)who were hospitalized in the obstetrics and gynaecology department of Wanbei Coal Power Group General Hospital were collected.The apoptosis of trophoblast cells was detected by TUNEL method.The expression of Bcl-2 and Bax protein in trophoblast cells were detected by immunohistochemistry.Western blotting and real-time quantitative PCR(qPCR)were used respectively,to detect the expression of Bcl-2 and Bax protein and mRNA.Apoptotic nuclei are mainly found in syncytiotrophoblasts,while is rarely found in cell trophoblasts,and the frequency of apoptosis is negatively correlated with gestational age.Bcl-2 and Bax proteins expressed in Syncytiotrophoblast in all gestational ages,while not in trophoblasts or other parts.The expression levels of Bcl-2 and Bax proteins were positively correlated with gestational age,and the expressions of Bcl-2 and Bax proteins in the full-term placenta were significantly higher than those in the first-term placenta(p<0.05)and mid-term placenta(p<0.05).The expression levels of Bcl-2 mRNA and Bax m RNA were positively correlated with gestational age,and mRNA expression levels were significantly different between first,mid and full-term gestation(p<0.05).Bcl-2 and Bax may play an important role in regulating the apoptosis of trophoblast cells,especially syncytiotrophoblasts.The mechanism may be to play a role in resisting apoptosis in the term placenta,so as to maintain the number of trophoblast cells in term placenta and ensure normal placental function.
