摘 要:中药抗炎治疗成为治疗乳腺炎的有效方法。厚朴酚(Magnolol)是从厚朴树皮中提取的一种多酚联萘化合物,已被证明具有潜在的抗炎活性。本研究旨在探讨厚朴酚对脂多糖(LPS)诱导的小鼠乳腺炎模型体内炎症的保护作用及其对脂多糖(LPS)刺激的小鼠乳腺上皮细胞(MMECs)的保护作用机制。使用组织病理学和髓过氧化物酶(MPO)测定组织损伤程度。酶联免疫吸附试验(ELISA)检测促炎细胞因子的表达。Western blotting检测核因子-κB(NF-κB)、抑制性κB(IκBα)蛋白、p38、细胞外信号调节激酶、c-Jun氨基末端激酶的表达。结果表明,厚朴酚在体内和体外均能明显抑制脂多糖诱导的TNF-α、IL-6和IL-1β的产生。厚朴酚可降低LPS刺激的内皮细胞I-κBα、p65、p38、ERK和JNK的磷酸化水平。在体内实验中,还观察到厚朴酚对小鼠乳房炎组织的损伤作用。这些结果表明,厚朴酚通过抑制NF-κB/丝裂原活化蛋白激酶信号系统而减轻LPS刺激的炎症反应。说明厚朴酚可能是一种治疗乳腺炎的药物。Anti-inflammatory treatment with traditional Chinese medicine has become an effective method to treat mastitis.Magnolol is a polyphenol-linked naphthalene compound extracted from bark of Magnolia officinalis and has been shown to have potential anti-inflammatory activity.The aim of this study was to investigate the protective effect of magnolol on inflammation in mouse mastitis induced by lipopolysaccharide(LPS)and the protective mechanism of magnolol on mouse mammary epithelial cells(MMECs)stimulated by lipopolysaccharide(LPS).Histopathology and myeloperoxidase(MPO)were used to determine the extent of tissue damage.Enzyme-linked immunosorbent assay(ELISA)was used to detect the expression of pro-inflammatory cytokines.Western blot was used to detect the expression of nuclear factor-κB(NF-κB),inhibitory-κB(IκBαprotein),p38,extracellular signal-regulated kinase,and c-Jun amino-terminal kinase.The results showed that magnolol significantly inhibited the production of tumor necrosis factor-α,interleukin-6 and interleukin-1 induced by LPS in vivo and in vitro.Honokiol decreased the phosphorylation of I-κBα,p65,p38,ERK and JNK in lps-stimulated endothelial cells.In vivo experiments,the damage effect of magnolol on mastitis tissues in mice was also observed.These results suggest that magnolol reduces the inflammatory response to LPS stimulation by inhibiting the signaling system of NF-κB/mitogen-activated protein kinase.This suggests that magnolol may be a medicine to treat mastitis.
